Apparatuses, systems and methods for imaging flow cytometry
The present disclosure provides apparatuses, systems, and methods for performing particle analysis through flow cytometry at comparatively high event rates and for gathering high resolution images of particles.
1 . A method, comprising:
selectively sorting at least some of a population of particles into a sorted particle subpopulation,
the selective sorting being performed on the basis of comparing a record associated with a given particle to predetermined gating conditions; and
recording an image of at least a particle sorted into the sorted particle subpopulation.
2 . The method of claim 1 , wherein the record is indicative of one or more of a cellular marker, a cellular function, or a physical characteristic of the given particle.
3 . The method of claim 1 , wherein the record associated with a given particle is obtained by illuminating the particle.
4 . The method of claim 1 , wherein the sorted subpopulation comprises particles having records that satisfy all predetermined gating conditions.
5 . The method of claim 1 , wherein the sorted subpopulation comprises particles having records that satisfy fewer than all predetermined gating conditions.
6 . The method of claim 1 , comprising recording only images of particles sorted into the sorted subpopulation.
7 . The method of claim 1 , wherein the population of particles is communicated in a flow path.
8 . A method, comprising:
selectively recording an image of at least one particle within an imaged particle subpopulation of particles,
the selective recording being performed on the basis of comparing a record associated with the at least one particle to predetermined gating conditions;
and sorting the at least one particle into a sorted particle subpopulation.
9 . The method of claim 8 , wherein the record is indicative of one or more of a cellular marker, a cellular function, or a physical characteristic of the at least one particle.
10 . The method of claim 8 , wherein the record associated with a given particle is obtained by illuminating the particle.
11 . The method of claim 8 , wherein the imaged subpopulation comprises particles having records that satisfy all predetermined gating conditions.
12 . The method of claim 8 , wherein the imaged subpopulation comprises particles having records that satisfy fewer than all predetermined gating conditions.
13 . The method of claim 8 , comprising recording only images of particles within the imaged particle subpopulation of particles.
14 . The method of claim 8 , wherein the imaged particle subpopulation of particles is communicated in a flow path.
15 . A system, comprising:
a flow path configured to communicate particles therethrough;
a first train,
the first train configured to illuminate at least one particle at a location along the flow path and the first train being configured to derive for the at least one particle a record associated with illumination of the at least one particle by the first train; and
a second train,
the second train being configured to selectively sort particles into a sorted particle subpopulation on the basis of comparing the record of a particle to predetermined gating conditions and the second train being configured to record an image of a particle of the sorted particle subpopulation.
16 . The system of claim 15 , wherein the record is indicative of one or more of a cellular marker, a cellular function, or a physical characteristic of the at least one particle.
17 . The system of claim 15 , wherein the second train is configured to record only images of particles sorted into the sorted particle subpopulation.
18 . A system, comprising:
a first flow path configured to communicate particles therethrough;
a first train,
the first train configured to illuminate at least one particle at a location along the flow path and the first train configured to derive for the at least one particle a record associated with illumination of the at least one particle by the first train; and
a second train,
the second train being configured to selectively sort particles into a sorted particle subpopulation of particles, the selective sorting being performed on the basis of comparing the record associated with a particle to predetermined gating conditions, and the second train being configured to move particles of the sorted particle subpopulation into any one or more of a second flow path, a chamber, or a region of a flow in the first flow path that has a speed different than a speed at the location along the flow path.
19 . The system of claim 18 , wherein the record is indicative of one or more of a cellular marker, a cellular function, or a physical characteristic of the at least one particle.
20 . The system of claim 18 , wherein the second train is configured to record only images of particles sorted into the sorted particle subpopulation.
