Abstract

Disclosed in the present invention are a compound shown in formula (IA), an optical isomer thereof and a pharmaceutically acceptable salt thereof, and the use of the compound as a STING agonist.

Claims (63)

1 . A compound of formula (IA), an optical isomer thereof or a pharmaceutically acceptable salt thereof,

wherein,

L 1 is selected from the group consisting of —O—, —NH— and a single bond;

R 1 is selected from the group consisting of H and C 1-6 alkyl, wherein the C 1-6 alkyl is optionally substituted with 1, 2 or 3 R;

R 2 and R 3 are each independently selected from the group consisting of H,

C 3-6 cycloalkyl, 5-6 membered heterocycloalkyl and 5-6 membered heteroaryl, wherein the C 3-6 cycloalkyl, 5-6 membered heterocycloalkyl or 5-6 membered heteroaryl is optionally substituted with 1, 2 or 3 R;

L 2 is selected from the group consisting of a single bond, —O—, —S—, —NH— and —NHC(═O)—;

R 4 is selected from the group consisting of

m is selected from the group consisting of 1, 2, 3, 4, 5 and 6;

R 5 is independently selected from the group consisting of OH, NH 2 , —CH 2 NH 2 ,

R 6 and R 7 are included in each instance of repeating unit of variable m, and in each instance,

R 6 and R 7 are each independently selected from the group consisting of H, halogen, N 3 , OH, SH, NH 2 , CN,

C 1-6 alkyl, C 1-6 alkoxy, C 1-6 alkylthio and C 1-6 alkylamino, wherein the C 1-6 alkyl, C 1-6 alkoxy, C 1-6 alkylthio or C 1-6 alkylamino is optionally substituted with 1, 2 or 3 R;

R 8 is selected from the group consisting of H, halogen, N 3 , OH, SH, NH 2 , CN,

and C 1-6 alkyl, wherein the C 1-6 alkyl is optionally substituted with 1, 2 or 3 R;

or, R 3 and R 8 are linked to form a 5-6 membered heterocyclic ring;

R 9 and R 12 are each independently selected from the group consisting of H and C 1-6 alkyl, wherein the C 1-6 alkyl is optionally substituted with 1, 2 or 3 R;

R 10 , R 11 , R 13 and R 14 are each independently selected from the group consisting of H, halogen, N 3 , OH, SH, NH 2 , CN and C 1-6 alkyl, wherein the C 1-6 alkyl is optionally substituted with 1, 2 or 3 R;

or, R 9 and R 13 are linked to form a carbon chain comprising 3-7 carbon atoms;

T is selected from the group consisting of N and CH;

R is independently selected from the group consisting of H, halogen, N 3 , OH, SH, NH 2 , CN,

C 1-6 alkyl, C 1-6 alkoxy, C 1-6 alkylthio, C 1-6 alkylamino, 5-6 membered heterocycloalkyl, phenyl and 5-10 membered heteroaryl, wherein the C 1-6 alkyl, C 1-6 alkoxy, C 1-6 alkylthio, C 1-6 alkylamino, 5-6 membered heterocycloalkyl, phenyl or 5-10 membered heteroaryl is optionally substituted with 1, 2 or 3 R′;

R′ is selected from the group consisting of H, F, Cl, Br, I, OH, NH 2 and CH 3 ;

the 5-6 membered heterocycloalkyl, 5-6 membered heteroaryl, 4-10 membered heterocycloalkyl or 5-10 membered heteroaryl contains 1, 2 or 3 heteroatoms or heteroatom groups independently selected from the group consisting of —O—, —NH—, —S— and N.

2 . The compound, the optical isomer thereof or the pharmaceutically acceptable salt thereof according to claim 1 , selected from:

R 1 is selected from the group consisting of H and C 1-6 alkyl, wherein the C 1-6 alkyl is optionally substituted with 1, 2 or 3 R;

R 2 and R 3 are each independently selected from the group consisting of H,

C 3-6 cycloalkyl, 5-6 membered heterocycloalkyl and 5-6 membered heteroaryl, wherein the C 3-6 cycloalkyl, 5-6 membered heterocycloalkyl or 5-6 membered heteroaryl is optionally substituted with 1, 2 or 3 R;

L 2 is selected from the group consisting of a single bond, —O—, —S—, —NH— and —NHC(═O)—;

R 4 is selected from the group consisting of

m is selected from the group consisting of 1, 2, 3, 4, 5 and 6;

R 5 is independently selected from the group consisting of OH, NH 2 , —CH 2 NH 2 ,

R 6 and R 7 are included in each instance of repeating unit of variable m, and in each instance,

R 6 and R 7 are each independently selected from the group consisting of H, halogen, N 3 , OH, SH, NH 2 , CN,

C 1-6 alkyl, C 1-6 alkoxy, C 1-6 alkylthio and C 1-6 alkylamino, wherein the C 1-6 alkyl, C 1-6 alkoxy, C 1-6 alkylthio or C 1-6 alkylamino is optionally substituted with 1, 2 or 3 R;

R 9 is independently selected from the group consisting of H and C 1-6 alkyl, wherein the C 1-6 alkyl is optionally substituted with 1, 2 or 3 R;

R 13 is independently selected from the group consisting of H, halogen, N 3 , OH, SH, NH 2 , CN and C 1-6 alkyl, wherein the C 1-6 alkyl is optionally substituted with 1, 2 or 3 R;

or, R 9 and R 13 are linked to form a carbon chain comprising 3-7 carbon atoms;

R is independently selected from the group consisting of H, halogen, N 3 , OH, SH, NH 2 , CN,

C 1-6 alkyl, C 1-6 alkoxy, C 1-6 alkylthio, C 1-6 alkylamino, 5-6 membered heterocycloalkyl, phenyl and 5-10 membered heteroaryl, wherein the C 1-6 alkyl, C 1-6 alkoxy, C 1-6 alkylthio, C 1-6 alkylamino, 5-6 membered heterocycloalkyl, phenyl or 5-10 membered heteroaryl is optionally substituted with 1, 2 or 3 R′;

R′ is selected from the group consisting of H, F, Cl, Br, I, OH, NH 2 and CH 3 ;

the 5-6 membered heterocycloalkyl, 5-6 membered heteroaryl, 5-10 membered heterocycloalkyl or 5-10 membered heteroaryl contains 1, 2 or 3 heteroatoms or heteroatom groups independently selected from the group consisting of —O—, —NH—, —S— and N.

3 . The compound, the optical isomer thereof or the pharmaceutically acceptable salt thereof according to claim 1 , wherein R is independently selected from the group consisting of H, halogen, N 3 , OH, SH, NH 2 , CN,

C 1-3 alkyl, C 1-3 alkoxy, C 1-3 alkylthio, C 1-3 alkylamino, morpholinyl, phenyl, imidazolyl and indolyl, wherein the C 1-3 alkyl, C 1-3 alkoxy, C 1-3 alkylthio, C 1-3 alkylamino, morpholinyl, phenyl, imidazolyl or indolyl is optionally substituted with 1, 2 or 3 R′.

4 . The compound, the optical isomer thereof or the pharmaceutically acceptable salt thereof according to claim 3 , wherein R is independently selected from the group consisting of H, F, Cl, Br, I, N 3 , OH, SH, NH 2 , CN, Me,

The compound, the optical isomer thereof or the pharmaceutically acceptable salt thereof according to claim 9 , wherein R 6 and R 7 are included in each instance of repeating unit of variable m, and in each instance, R 6 and R 7 are each independently selected from the group consisting of H, F, Cl, Br, I, N 3 , OH, SH, NH 2 , CN, Me,

13 . A pharmaceutical composition, comprising the compound or the pharmaceutically acceptable salt thereof according to claim 1 .

A method for the treatment of a STING-mediated disease or a STING-mediated tumor complication in a subject in need thereof, comprising administering the compound or the pharmaceutically acceptable salt thereof according to claim 1 to the subject, wherein the STING-mediated disease comprises a disease selected from the group consisting of cancer, inflammation, infectious diseases and immune-related diseases.

16 . The method according to claim 14 , wherein the STING-mediated tumor complication is selected from malignant pleural effusion and ascites.
