Abstract

Provided herein, in some embodiments, are engineered cells and use of the same for increased hydrogen production. In particular, provided herein are genetically engineered cells comprising a polynucleotide encoding a fusion protein comprising a photosystem I (PSI) protein and an algal hydrogenase, as well as methods for producing such genetically engineered cells. Also provided herein are methods for increasing hydrogen (H 2 ) production in cells.

Claims (4)

1 . A fusion protein comprising a photosystem I subunit (PsaC) protein and an algal hydrogenase, wherein the hydrogenase is inserted into the hinge region of the PsaC.

2 . The fusion protein of claim 1 , wherein the algal hydrogenase is hydrogenase A.

3 . The fusion protein of claim 2 , further comprising at least one amino acid at a junction between the PsaC and the N-terminal, the C-terminal, or both ends of the hydrogenase.

4 . The fusion protein of claim 1 , wherein the fusion protein comprises SEQ ID NO: 1, SEQ ID NO: 2 or a polypeptide having 95% identity to SEQ ID NO: 1 or SEQ ID NO: 2.
