Unglycosylated lysostaphin variant protein
Unglycosylated lysostaphin variant protein, nucleic acid molecule, vector and host cell, as well as a method for production of unglycosylated lysostaphin variant protein in a yeast expression system are provided. The proteins are produced in a Pichia pastoris expression system and have been shown to have activity equivalent to wild-type lysostaphin. The lysostaphin variant proteins can be used as therapeutic proteins for treatment of diseases such as Staphylococcus aureus infection.
1 . A method for inhibiting growth of bacteria, the method comprising contacting the bacteria with an effective amount of a lysostaphin (LST) variant protein comprising the amino acid sequence SEQ ID NO: 4 or SEQ ID NO: 6, wherein the contacting inhibits growth of the bacteria.
2 . The method of claim 1 , wherein the bacteria are Staphylococcus aureus ( S. aureus ) or methicillin-resistant S. aureus (MRSA).
3 . The method of claim 1 , wherein the LST variant protein is unglycosylated at the position corresponding to N125 of SEQ ID NO: 4.
4 . The method of claim 1 , wherein the LST variant protein is unglycosylated at the position corresponding to N125 of SEQ ID NO: 6.
5 . An isolated unglycosylated lysostaphin variant protein comprising the amino acid sequence of SEQ ID NO: 4 or SEQ ID NO: 6.
6 . The isolated unglycosylated lysostaphin variant protein of claim 5 , wherein said protein comprises the amino acid sequence of SEQ ID NO: 4.
7 . The isolated unglycosylated lysostaphin variant protein of claim 6 , which is unglycosylated at the position corresponding to N125 of SEQ ID NO: 4.
8 . The isolated unglycosylated lysostaphin variant protein of claim 5 , wherein said protein comprises the amino acid sequence of SEQ ID NO: 6.
9 . The isolated unglycosylated lysostaphin variant protein of claim 8 , which is unglycosylated at the position corresponding to N125 of SEQ ID NO: 6.
