Abstract

A method for promoting diversification of variable regions of an antibody, particularly a method for promoting diversification of the amino acid sequences of variable regions of an antibody generated by an avian B cell population, the method including suppressing the PI3Kα activity of each avian B cell comprised in the avian B cell population expressing the antibody.

Claims (5)

1 . A method for promoting diversification of amino acid sequences of variable regions of an antibody generated by an avian B cell population, wherein the method comprises:

contacting a population of avian B cells with a PI3Kα inhibitor to diversify amino acid sequences of variable regions of an antibody generated by said avian B cell population; and

sequencing amino acid sequences of variable regions of the antibodies generated by said avian B cell population after said contact with said PI3Kα inhibitor to confirm diversification of amino acid sequences of variable regions of said antibodies generated by said avian B cell population after contact with said PI3Kα inhibitor.

2 . A method for producing an antibody comprising performing the method of claim 1 and isolating an antibody having a diversification of amino acid sequences of variable regions after said sequencing.

3 . The method of claim 1 , wherein the PI3Kα inhibitor is LY294002.
