Abstract

Disclosed are IgG2 constant regions and human κ chain constant regions with altered amino acid sequences, antibodies comprising these constant regions, and pharmaceutical compositions comprising the antibodies.

Claims (27)

1 . An antibody constant region comprising the amino acid sequence of SEQ ID NO: 24 with at least the following substitutions and optional deletions:

Ser is substituted for Cys at position 14, which corresponds to EU numbering position 131;

Lys is substituted for Arg at position 16, which corresponds to EU numbering position 133;

Ser is substituted for Cys at position 103, which corresponds to EU numbering position 220;

Gly is substituted for Glu at position 20, which corresponds to EU numbering position 137;

Gly is substituted for Ser at position 21, which corresponds to EU numbering position 138;

Gln is substituted for His at position 147, which corresponds to EU numbering position 268;

Gln is substituted for Arg at position 234, which corresponds to EU numbering position 355; and

Glu is substituted for Gln at position 298, which corresponds to EU numbering position 419,

wherein Gly at position 325 and Lys at position 326 of SEQ ID NO: 24, corresponding to EU numbering positions 446 and 447, respectively, are optionally deleted.

2 . An antibody constant region comprising the amino acid sequence of SEQ ID NO: 24 with at least the following substitutions and optional deletions:

Ser is substituted for Cys at position 14, which corresponds to EU numbering position 131;

Lys is substituted for Arg at position 16, which corresponds to EU numbering position 133;

Ser is substituted for Cys at position 103, which corresponds to EU numbering position 220;

Gly is substituted for Glu at position 20, which corresponds to EU numbering position 137;

Gly is substituted for Ser at position 21, which corresponds to EU numbering position 138;

Gln is substituted for His at position 147, which corresponds to EU numbering position 268;

Gln is substituted for Arg at position 234, which corresponds to EU numbering position 355;

Glu is substituted for Gln at position 298, which corresponds to EU numbering position 419;

Ser is substituted for Ala at position 209, which corresponds to EU numbering position 330;

Ser is substituted for Pro at position 210, which corresponds to EU numbering position 331; and

Ala is substituted for Thr at position 218, which corresponds to EU numbering position 339,

wherein Gly at position 325 and Lys at position 326 of SEQ ID NO: 24, corresponding to EU numbering positions 446 and 447, respectively, are optionally deleted.

3 . The antibody constant region of claim 1 , wherein Gly at position 325 and Lys at position 326 of SEQ ID NO: 24 are deleted.

4 . The antibody constant region of claim 2 , wherein Gly at position 325 and Lys at position 326 of SEQ ID NO: 24 are deleted.

5 . An antibody comprising the constant region of any one of claims 1, 2, 3, and 4 .

6 . A pharmaceutical composition comprising the antibody of claim 5 .
