Abstract

The present disclosure provides methods to genetically engineer plants by manipulating the expression of the PtrXB38 polypeptide to increase root development. Also provided are genetically engineer plants that can be obtained by the methods according to the present disclosure.

Claims (39)

1 . A genetically modified plant, plant cell or plant tissue, comprising an exogenous nucleic acid which comprises a nucleotide sequence encoding a PtrXB38 polypeptide,

wherein (i) the nucleotide sequence has at least 98% sequence identity to the nucleotide sequence shown in SEQ ID NO: 1, or (ii) the PtrXB38 polypeptide has at least 98% sequence identity to the amino acid sequence shown in SEQ ID NO: 2, and

wherein the genetically modified plant, plant cell or plant tissue displays an enhanced expression of the PtrXB38 polypeptide and an increase in root development compared to a wild-type plant, plant cell, or plant tissue.

2 . The genetically modified plant, plant cell or plant tissue of claim 1 , wherein the exogenous nucleic acid is stably integrated into the plant genome.

3 . The genetically modified plant, plant cell or plant tissue of claim 1 , wherein the nucleotide sequence is operably linked to a heterologous promoter.

4 . The genetically modified plant, plant cell or plant tissue of claim 3 , wherein the heterologous promoter is a constitutive promoter, an inducible promoter, or a native promoter.

5 . The genetically modified plant, plant cell or plant tissue of claim 3 , wherein the heterologous promoter is a 35S promoter.

6 . The genetically modified plant, plant cell or plant tissue of claim 1 , wherein the plant is a monocot or a dicot.

7 . The genetically modified plant, plant cell or plant tissue of claim 6 , wherein the plant is selected from the group consisting of genera Eucalyptus, Populus , and Salix.

8 . The genetically modified plant, plant cell or plant tissue of claim 7 , wherein the plant is Populus.

9 . The genetically modified plant, plant cell or plant tissue of claim 8 , wherein the plant is Populus trichocarpa.

10 . The genetically modified plant, plant cell or plant tissue of claim 1 , wherein the root development comprises adventitious root formation.

11 . The genetically modified plant, plant cell or plant tissue of claim 10 , wherein the adventitious root formation requires auxin transport and signaling.

12 . The genetically modified plant, plant cell or plant tissue of claim 10 , wherein the adventitious root formation requires ethylene biosynthesis and signaling.

13 . The genetically modified plant, plant cell or plant tissue of claim 1 , wherein the enhanced expression of the PtrXB38 polypeptide increases flavonoids in roots.

14 . A method of increasing root development in a plant, plant cell or plant tissue, comprising introducing into the plant, plant cell or plant tissue, an exogenous nucleic acid sequence encoding a PtrXB38 polypeptide, thereby obtaining a genetically modified plant, plant cell or plant tissue,

wherein (i) the nucleotide sequence has at least 98% sequence identity to the nucleotide sequence shown in SEQ ID NO: 1, or (ii) the PtrXB38 polypeptide has at least 98% sequence identity to the amino acid sequence shown in SEQ ID NO: 2, and

wherein the genetically modified plant, plant cell or plant tissue displays an enhanced expression of the PtrXB38 polypeptide and an increase in root development compared to a wild-type plant, plant cell, or plant tissue.

15 . The method of claim 14 , wherein the exogenous nucleic acid is stably integrated into the plant genome.

16 . The method of claim 14 , wherein the nucleotide sequence is operably linked to a heterologous promoter.

17 . The method of claim 16 , wherein the heterologous promoter is a constitutive promoter or an inducible promoter.

18 . The method of claim 16 , wherein the heterologous promoter is a 35S promoter.

19 . The method of claim 14 , wherein the plant is a monocot or a dicot.

20 . The method of claim 19 , wherein the plant is selected from the group consisting of genera Eucalyptus, Populus , and Salix.

21 . The method of claim 20 , wherein the plant is Populus.

22 . The method of claim 21 , wherein the plant is Populus trichocarpa.

23 . The method of claim 14 , wherein the root development comprises adventitious root formation.

24 . The method of claim 23 , wherein the adventitious root formation requires auxin transport and signaling.

25 . The method of claim 23 , wherein the adventitious root formation requires ethylene biosynthesis and signaling.

26 . The method of claim 23 , wherein the enhanced expression of the PtrXB38 polypeptide increases flavonoids in roots.

27 . A method of increasing flooding tolerance in a plant or plant tissue comprising enhancing expression of a PtrXB38 polypeptide in the plant, or plant tissue, wherein the PtrXB38 polypeptide has at least 98% sequence identity to the amino acid sequence shown in SEQ ID NO: 2, thereby increasing flooding tolerance in the plant or plant tissue.

28 . The method of claim 27 , wherein the exogenous nucleic acid is stably integrated into the plant genome.

29 . The method of claim 27 , wherein the nucleotide sequence is operably linked to a heterologous promoter.

30 . The method of claim 29 , wherein the heterologous promoter is a constitutive promoter, or an inducible promoter.

31 . The method of claim 29 , wherein the heterologous promoter is a 35S promoter.

32 . The method of claim 27 , wherein the plant is a monocot or a dicot.

33 . The method of claim 32 , wherein the plant is selected from the group consisting of genera Eucalyptus, Populus , and Salix.

34 . The method of claim 33 , wherein the plant is Populus.

35 . The method of claim 34 , wherein the plant is Populus trichocarpa.
