Abstract

The invention relates to a complex comprising an antibiotic substance and a nucleophilic derivatization reagent, compositions comprising the complex, kits comprising complex or composition, as well as uses of the complex or composition.

Claims (17)

1 . A complex formed of an antibiotic substance and a nucleophilic derivatization reagent, the complex including an isotopic label, wherein the nucleophilic derivatization reagent is a linear primary amine comprising a primary amine group and in the range of from 3 to 10 C-atoms, and wherein the antibiotic substance is a beta-lactam antibiotic.

2 . The complex of claim 1 , wherein the antibiotic substance is Meropenem or Piperacillin.

3 . A composition comprising the complex of claim 1 .

4 . A composition comprising the complex of claim 1 , wherein the composition further comprises a solvent.

5 . The composition of claim 4 , further comprising a non-nucleophilic base that is stable and miscible with water.

6 . A kit comprising a) the complex of claim 1 , and b) a package insert.

A derivatized beta-lactam antibiotic substance comprising an isotopic label, derivatized with a nucleophilic derivatization reagent, wherein the nucleophilic derivatization reagent is a linear primary amine comprising a primary amine group and in the range of from 3 to 10 C-atoms, the derivatized beta-lactam antibiotic substance being suitable as an internal standard (ISTD) and/or a calibrator in Mass Spectrometry Measurements.

A method of producing a stabilized antibiotic substance, comprising derivatizing a beta-lactam antibiotic substance with a nucleophilic derivatization reagent to stabilize the antibiotic substance, wherein the nucleophilic derivatization reagent is a linear primary amine comprising a primary amine group and in the range of from 3 to 10 C-atoms, wherein the antibiotic substance and/or the nucleophilic derivatization reagent comprises one or more isotopic label.

The composition of claim 4 , wherein the solvent is selected from the group consisting of water, CH 3 CN, THF, Dioxanes, DMF, DMSO, acetone, t-butyl alcohol, diglyme, DME, MeOH, EtOH, 1-PrOH, 2-PrOH, ethylene glycol, Hexamethylphosphoramiede (HMPA), Hexamethylphosphorous triamide (HMPT), and glycerin.

10 . The composition of claim 5 , wherein the non-nucleophilic base is selected from the group consisting of DBU, Na 3 PO 4 , Na 2 CO 3 , and Cs 2 CO 3 .

11 . The method of claim 8 , wherein the antibiotic substance is Meropenem or Piperacillin.

12 . The method of claim 8 , wherein the nucleophilic derivatization reagent stabilizes the antibiotic substance by preventing hydrolyzation of the antibiotic substance.

13 . The method of claim 8 , wherein the nucleophilic derivatization reagent stabilizes the antibiotic substance for more than 12 hours.

14 . The method of claim 8 , wherein the nucleophilic derivatization reagent is provided in an excess of from about 1E5 to about 1E10 compared to the amount of antibiotic substance present.

15 . The method of claim 8 , wherein the derivatized antibiotic substance is used as an internal standard (ISTD) and/or a calibrator in Mass Spectrometry Measurements.

16 . The complex of claim 1 , wherein the complex comprises one or more isotopic label(s) in the antibiotic substance of the complex.

17 . The complex of claim 1 , wherein the complex comprises 1 to 10 isotopic label(s) in the antibiotic substance of the complex.
