Abstract

Among the various aspects of the present disclosure is the provision of methods of treating cancer in a subject comprising administering a TYK2 inhibiting agent and a MEK inhibiting agent. In some embodiments, the subject has a malignant peripheral nerve sheath tumor (MPNST), neurofibromatosis Type 1 (NF1) cancer predisposition syndrome, benign plexiform neurofibromas (PN), or atypical neurofibromas (ANNUBP). Methods of inhibiting TYK2 and MEK in a subject in need thereof are also provided.

Claims (4)

1 . A method of treating cancer comprising administering a synergistic amount of a TYK2 inhibiting agent and an MEK inhibiting agent, wherein the TYTK2 inhibiting agent comprises at least one of deucravacitinib and WU-12 and the MEK inhibiting agent comprises mirdametinib; and wherein the cancer is malignant peripheral nerve sheath tumor (MPNST).

2 . The method of claim 1 , wherein the MPNST overexpresses TYK2 or comprises a TYK2 mutation.

3 . The method of claim 1 , wherein the subject has neurofibromatosis Type 1 (NF1) cancer predisposition syndrome, benign plexiform neurofibromas (PN), or atypical neurofibromas (ANNUBP).

4 . The method of claim 1 , wherein the TYK2 inhibiting agent does not inhibit JAK1, JAK2, or JAK3.
