Abstract

Among other things, disclosed herein are compositions and methods for using human-isolated or synthetic Alcaligenes faecalis, Bacillus altitudinis, Bacillus pumilus, Bacillus subtilis , or Janthinobacterium lividum . Compositions may improve skin health. Methods may include applying human-isolated or synthetic Alcaligenes faecalis, Bacillus altitudinis, Bacillus pumilus, Bacillus subtilis , or Janthinobacterium lividum over a host or host area, such as skin or mucosa to minimize the presence of one or more pathogenic microbes, maximize therapeutic effects of one or more additional therapeutics, and/or improve health of a host or host area.

Claims (19)

1 . A composition comprising a probiotic, wherein the probiotic comprises human-isolated or synthetic Alcaligenes faecalis in an amount effective to treat a disease, disorder, or condition associated with a pathogenic microorganism, and a cryoprotectant, wherein the probiotic comprises a nucleic acid sequence at least 95% identical to that of SEQ ID NO: 6.

2 . The composition of claim 1 , formulated for topical application to a mammal for treating the disease, disorder, or condition, wherein the disease disorder, or condition is an infection.

3 . The composition of claim 2 , wherein the cryoprotectant results in greater efficacy, stability, and/or viability of the human-isolated or synthetic Alcaligenes faecalis as compared to a composition comprising the same human-isolated or synthetic Alcaligenes faecalis without the cryoprotectant.

4 . The composition of claim 1 , wherein the composition is frozen or lyophilized.

5 . The composition of claim 1 , wherein the cryoprotectant results in a greater percent recovery of the human-isolated or synthetic Alcaligenes faecalis after freezing or lyophilization as compared to a composition comprising the same human-isolated or synthetic Alcaligenes faecalis without the cryoprotectant.

6 . The composition of claim 1 , wherein the cryoprotectant comprises a disaccharide.

7 . The composition of claim 6 , wherein the disaccharide comprises trehalose.

8 . The composition of claim 7 , wherein the trehalose comprises D-trehalose, wherein the D-trehalose is about 2% to about 20% of the composition by weight.

9 . The composition of claim 1 , further comprising at least one additional isolated or synthetic microbe.

10 . The composition of claim 9 , wherein the additional isolated or synthetic microbe comprises a human-isolated or synthetic Bacillus altitudinis, Bacillus pumilus, Bacillus subtilis , or Janthinobacterium lividum.

11 . The composition of claim 9 , wherein the additional isolated or synthetic microbe comprises one or more microbes selected from Lactobacillus, Lactococcus, Cutibacterium , and Propionibacterium.

12 . The composition of claim 1 , further comprising a first additional isolated microbe and a second additional isolated microbe, wherein the first and second additional isolated microbes are independently selected from bacteria, virus, yeast, fungus, and any combination thereof.

13 . The composition of claim 1 , wherein:

a) the disease, disorder, or condition is associated with Malassezia, Staphylococcus, Candida , or Trichophyton ; and/or

b) the disease, disorder, or condition is Tinea barbae, Tinea capitis, Tinea corporis, Tinea cruris, Tinea pedis, Tinea versicolor , or onychomycosis or any combination thereof.

14 . The composition of claim 1 , further comprising a metabolite, cell lysate, or postbiotic from at least one species of human-isolated or synthetic bacteria selected from Alcaligenes faecalis, Bacillus altitudinis, Bacillus pumilus , and Bacillus subtilis.

15 . A probiotic composition comprising Alcaligenes faecalis and at least one cryoprotectant for use as a medicament, wherein the probiotic comprises a nucleic acid sequence at least 95% identical to that of SEQ ID NO: 6.

16 . The probiotic composition of claim 15 , for use in the treatment of a disease, disorder, or condition associated with at least one pathogenic microorganism.

17 . A kit comprising (i) at least one vial comprising the composition of claim 1 , and (ii) instructions.
