Human immunodeficiency virus (HIV) nucleotide sequences, recombinant polypeptides, and applications thereof
View Patent ↗Polynucleotide sequences are provided for the diagnosis of the presence of retroviral infection in a human host associated with lymphadenopathy syndrome and/or acquired immune deficiency syndrome, for expression of polypeptides and use of the polypeptides to prepare antibodies, where both the polypeptides and antibodies may be employed as diagnostic reagents or in therapy, e.g., vaccines and passive immunization. The sequences provide detection of the viral infectious agents associated with the indicated syndromes and can be used for expression of antigenic polypeptides.
1. A method for replicating DNA specific for HIV, which comprises:
(a) providing a DNA construct comprising an origin of replication recognized by a unicellular microorganism and a DNA sequence comprising at least a 20 bp sequence of a human immunodeficiency virus (HIV) genome; and
(b) growing a unicellular microorganism containing said DNA construct under conditions whereby said DNA sequence is replicated.
2. The method according to claim 1 wherein the DNA construct contains a gag sequence of HIV.
3. The method according to claim 1 wherein the DNA construct contains a env sequence of HIV.
4. The method according to claim 1 wherein the DNA construct contains an HIV pol sequence having a nucleic acid sequence between the BstXI site at position 3006 and the NdeI site at position 5131 as set forth in FIG. 4 .
5. The method of claim 1 , 2 or 3 wherein the unicellular microorganism is a eukaryotic cell.
6. The method of claim 1 , 2 or 3 wherein the unicellular microorganism is a yeast cell.
7. The method of claim 1 , 2 or 3 wherein the unicellular microorganism is a bacterial cell.
8. The method of claim 1 , 2 , or 3 herein the HIV sequence is a gag sequence of FIG. 4 .
9. The method of claim 1 , 2 , or 3 wherein the HIV sequence is an env sequence of FIG. 4 .
10. The method of claim 1 , 2 , or 3 wherein the HIV sequence is at least 45 base pairs long.
11. A method for replicating DNA specific for HIV, which comprises:
(a) providing a DNA construct comprising an origin of replication recognized by a unicellular microorganism and a DNA sequence comprising at least a 20 basepair sequence of a human immunodeficiency virus (HIV) pol sequence as shown between the BstXI site at position 3006 and the NdeI site at position 5131 in FIG. 4 ; and
(b) growing a unicellular microorganism containing the DNA construct under conditions whereby the DNA sequence is replicated.
12. The method of claim 11 wherein the DNA sequence comprises at least 50 basepairs.
13. The method of claim 11 wherein the DNA sequence comprises at least 100 basepairs.
14. A method for replicating DNA specific for HIV, which comprises:
(a) providing a DNA construct comprising an origin of replication recognized by a unicellular microorganism and a DNA sequence comprising at least a 20 basepair sequence of a human immunodeficiency virus (HIV) gag sequence as shown in FIG. 4 ; and
(b) growing a unicellular microorganism containing the DNA construct under conditions whereby the DNA sequence is replicated.
15. The method of claim 14 wherein the DNA sequence comprises at least 50 basepairs.
16. The method of claim 14 wherein the DNA sequence comprises at least 100 basepairs.
17. A method for replicating DNA specific for HIV, which comprises:
(a) providing a DNA construct comprising an origin of replication recognized by a unicellular microorganism and a DNA sequence comprising at least a 20 basepair sequence of a human immunodeficiency virus (HIV) env sequence as shown in FIG. 4 ; and
(b) growing a unicellular microorganism containing the DNA construct under conditions whereby the DNA sequence is replicated.
18. The method of claim 17 wherein the DNA sequence comprises at least 50 basepairs.
19. The method of claim 17 wherein the DNA sequence comprises at least 100 basepairs.
