Abstract

The present invention relates to monoclonal antibodies that bind to the extracellular domain of prostate-specific membrane antigen (PSMA), hybridoma cell lines producing the antibodies, and methods of using such antibodies for diagnosis and treatment of cancer. In particular, thirty-five monoclonal antibodies reactive with PSMA expressed on the cell surface are exemplified. Additionally, the present invention relates to a novel protein variant (PSM′) of PSMA detected by a number of the antibodies of the invention. The hydrolase activity of PSMA and PSM′ allows the use of an immunoenzymatic assay for their detection.

Claims (24)

1. A method of detecting the presence of prostate-specific membrane antigen (PSMA) expressed by prostate cancer cells, comprising contacting a sample of cells in vivo with a first labeled monoclonal antibody having an antigen-binding region binding to an epitope in the extracellular domain of PSMA, and detecting any antibody-bound PSMA on the membrane of the prostate cancer cells, in which the antigen-binding region of the first labeled antibody is capable of competitively inhibiting the immunospecific binding of a second monoclonal antibody to its target epitope, wherein said second antibody is produced by hybridoma 3F5.4G6 having ATCC accession number HB 12060, 3D7-1.1 having ATCC accession number HB 12309, 4E10-1.14 having ATCC accession number HB 12310, 3C9 having ATCC accession number HB 12484, 2C7 having ATCC accession number HB 12490, 1G3 having ATCC accession number HB 12489, 4C8B9 having ATCC accession number HB 12492; 1G9 having ATCC accession number HB 12495, 3C6 having accession number HB 12491, or 4D4 having accession number HB 12493, thereby detecting the presence of prostate-specific membrane antigen (PSMA) expressed by prostate cancer cells.

2. The method of claim 1 in which the prostate cancer cells are metastatic prostate cancer cells.

3. The method of claim 1 in which the first labeled monoclonal antibody comprises a label which is a radioisotope.

The method of claim 1 , wherein the first labeled monoclonal antibody is produced by hybridoma 3F5.4G6 having ATCC accession number HB 12060, 3D7-1.1 having ATCC accession number HB 12309, 4E10-1.14 having ATCC accession number HB 12310, 3C9 having ATCC accession number HB 12484, 2C7 having ATCC accession number HB 12490, 1G3 having ATCC accession number HB 12489, 4C8B9 having ATCC accession number HB 12492, 1G9 having ATCC accession number HB 12495, 3C6 having accession number HB 12491, or 4D4 having accession number HB 12493.

5. The method of claim 1 , wherein the first labeled monoclonal antibody comprises a label which is a radiopaque dye.

6. The method of claim 1 , wherein the first labeled monoclonal antibody comprises a label which is a radioactive metal isotope.

7. The method of claim 1 , wherein the first labeled monoclonal antibody comprises a label which is a radioactive non-metal isotope.

8. The method of claim 1 , wherein the first labeled monoclonal antibody comprises a label which is a fluorogenic compound.

9. The method of claim 1 , wherein the first labeled monoclonal antibody comprises a label which is a fluorescent compound.

10. The method of claim 1 , wherein the first labeled monoclonal antibody comprises a label which is a positron emitting isotope.

11. The method of claim 1 , wherein the first labeled monoclonal antibody comprises a label which is a non-paramagnetic metal.

12. The method of claim 1 , wherein the second antibody is produced by hybridoma 3F5.4G6 having ATCC accession number HB 12060.

13. The method of claim 1 , wherein the second antibody is produced by hybridoma 3D7-1.1 having ATCC accession number HB 12309.

14. The method of claim 1 , wherein the second antibody is produced by hybridoma 4E10-1.14 having ATCC accession number HB 12310.

15. The method of claim 1 , wherein the second antibody is produced by hybridoma 3C9 having ATCC accession number HB 12484.

16. The method of claim 1 , wherein the second antibody is produced by hybridoma 2C7 having ATCC accession number HB 12490.

17. The method of claim 1 , wherein the second antibody is produced by hybridoma 1G3 having ATCC accession number HB 12489.

18. The method of claim 1 , wherein the second antibody is produced by hybridoma 4C8B9 having ATCC accession number HB 12492.

19. The method of claim 1 , wherein the second antibody is produced by hybridoma 1G9 having ATCC accession number HB 12495.

20. The method of claim 1 , wherein the second antibody is produced by hybridoma 3C6 having accession number HB 12491.

21. The method of claim 1 , wherein the second antibody is produced by hybridoma 4D4 having accession number HB 12493.

22. A method of detecting the presence of prostate-specific membrane antigen (PSMA) expressed by prostate cancer cells, comprising contacting a sample of cells in vivo with a first labeled monoclonal antibody having an antigen-binding region binding to an epitope in the extracellular domain of PSMA, and detecting any antibody-bound PSMA on the membrane of the prostate cancer cells, in which the antigen-binding region of the first labeled antibody is capable of competitively inhibiting the immunospecific binding of a second monoclonal antibody to its target epitope, wherein said second antibody is produced by hybridoma 3F5.4G6 having ATCC accession number HB 12060, 3D7-1.1 having ATCC accession number HB 12309, or 4E10-1.14 having ATCC accession number HB 12310, thereby detecting the presence of prostate-specific membrane antigen (PSMA) expressed by prostate cancer cells.

23. A method of detecting the presence of prostate-specific membrane antigen (PSMA) expressed by prostate cancer cells, comprising contacting a sample of cells in vivo with a first labeled monoclonal antibody having an antigen-binding region binding to an epitope in the extracellular domain of PSMA, and detecting any antibody-bound PSMA on the membrane of the prostate cancer cells, in which the antigen-binding region of the first labeled antibody is capable of competitively inhibiting the immunospecific binding of a second monoclonal antibody to its target epitope, wherein said second antibody is produced by hybridoma 3C9 having ATCC accession number HB 12484, 2C7 having ATCC accession number HB 12490, or 1G3 having ATCC accession number HB 12489, thereby detecting the presence of prostate-specific membrane antigen (PSMA) expressed by prostate cancer cells.

A method of detecting the presence of prostate-specific membrane antigen (PSMA) expressed by prostate cancer cells, comprising contacting a sample of cells in vivo with a first labeled monoclonal antibody having an antigen-binding region binding to an epitope in the extracellular domain of PSMA, and detecting any antibody-bound PSMA on the membrane of the prostate cancer cells, in which the antigen-binding region of the first labeled antibody is capable of competitively inhibiting the immunospecific binding of a second monoclonal antibody to its target epitope, wherein said second antibody is produced by hybridoma 4C8B9 having ATCC accession number HB 12492; 1G9 having ATCC accession number HB 12495, 3C6 having accession number HB 12491, or 4D4 having accession number HB 12493, thereby detecting the presence of prostate-specific membrane antigen (PSMA) expressed by prostate cancer cells.
