Abstract

The present invention is directed to novel polypeptides having sequence identity with IL-17 and to nucleic acid molecules encoding those polypeptides. Also provided herein are vectors and host cells comprising those nucleic acid sequences, chimeric polypeptide molecules comprising the polypeptides of the present invention fused to heterologous polypeptide sequences, antibodies which bind to the polypeptides of the present invention and to methods for producing the polypeptides of the present invention. Further provided herein are methods for treating degenerative cartilaginous disorders.

Claims (35)

1. An isolated polypeptide comprising at least 95% amino acid sequence identity to:

(a) the amino acid residues 1 to 197 of SEQ ID NO:3,

(b) the amino acid residues 19 to 197 of SEQ ID NO:3,

(c) the amino acids encoded by the full-length coding sequence of the cDNA deposited under ATCC accession number 203522, or

(d) the amino acids encoded by the full-length coding sequence of the cDNA deposited under ATCC accession number 203522 lacking its associated signal peptide encoding region;

wherein said isolated polypeptide is capable of inducing the production of TNF-α in human leukemia monocytic THP-1 cells.

2. The isolated polypeptide of claim 1 which comprises the amino acid residues 1 to 197 of SEQ ID NO:3.

3. The isolated polypeptide of claim 1 which comprises the amino acid residues 19 to 177 of SEQ ID NO:3.

4. The isolated polypeptide of claim 1 which comprises the amino acids encoded by the full-length coding sequence of the cDNA deposited under ATCC accession number 203522.

5. The isolated polypeptide of claim 1 which comprises the amino acids encoded by the full-length coding sequence of the cDNA deposited under ATCC accession number 203522 lacking its associated signal peptide encoding region.

6. A chimeric polypeptide comprising a polypeptide according to claim 1 fused to a heterologous polypeptide.

7. The chimeric polypeptide of claim 6 , wherein said heterologous polypeptide is an epitope tag or an Fc region of an immunoglobulin.

8. An isolated polypeptide comprising:

(a) amino acid residues 1 to 197 of SEQ ID NO:3,

(b) amino acid residues 19 to 197 of SEQ ID NO:3,

(c) the amino acids encoded by the full-length coding sequence of the cDNA deposited under ATCC accession number 203522, or

(d) the amino acids encoded by the full-length coding sequence of the cDNA deposited under ATCC accession number 203522 lacking its associated signal peptide encoding region.

9. The isolated polypeptide of claim 8 comprising amino acid residues 1 to 197 of SEQ ID NO:3.

10. The isolated polypeptide of claim 8 comprising amino acid residues 19 to 197 of SEQ ID NO:3.

11. The isolated polypeptide of claim 8 comprising the amino acids encoded by the full-length coding sequence of the cDNA deposited under ATCC accession number 203522.

12. The isolated polypeptide of claim 8 comprising the amino acids encoded by the full-length coding sequence of the cDNA deposited under ATCC accession number 203522 lacking its associated signal peptide encoding region.

13. A composition of matter comprising a polypeptide selected from the group consisting of:

(a) a PRO1122 polypeptide comprising amino acid residues 1 to 197 of SEQ ID NO:3, and

(b) a PRO1122 polypeptide comprising amino acid residues 19 to 197 of SEQ ID NO:3; in combination with a pharmaceutically acceptable carrier.

14. The composition of matter of claim 13 comprising a PRO1122 polypeptide comprising amino acid residues 1 to 197 of SEQ ID NO:3 in combination with a pharmaceutically acceptable carrier.

15. The composition of matter of claim 13 which comprises a PRO1122 polypeptide comprising amino acid residues 19 to 197 of SEQ ID NO:3 in combination with a pharmaceutically acceptable carrier.

16. An article of manufacturing comprising:

a container; and

a composition of matter comprising an isolated polypeptide having at least 95% amino acid sequence identity to:

(a) amino acid residues 1 to 197 of SEQ ID NO:3,

(b) amino acid residues 19 to 197 of SEQ ID NO:3,

(c) the amino acids encoded by the full-length coding sequence of the cDNA deposited under ATCC accession number 203522, Or

(d) the amino acids encoded by the full-length coding sequence of the cDNA deposited under ATCC accession number 203522 lacking its associated signal peptide encoding region;

wherein said isolated polypeptide is capable of inducing the production of TNF-α in human leukemia monocytic THP-1 cells.

17. The article of manufacture of claim 16 , further comprising a label affixed to said container, or a package insert included in said container, referring to the use of said composition for the treatment of a degenerative cartilagenous disorder in a mammal.
