Abstract

Polynucleotides encoding the IL-13 receptor and fragments thereof are disclosed. IL-13 receptor proteins, methods for their production, inhibitors of binding of IL-13 and its receptor and methods for their identification are also disclosed. Methods of medical treatment using such molecules and antagonists of the IL-13/IL-13R interaction are also provided.

Claims (24)

1. A method of reducing an allergen-induced airway hyper responsiveness, or an allergen-induced increase of mucus-containing cells in an airway epithelium, in a mammalian subject, said method comprising

administering to the subject a polypeptide encoded by a polynucleotide that hybridizes under conditions comprising hybridization at 65° C. in 0.1×SSC to the complement of the portion of SEQ ID NO:3 that encodes from amino acid 26 to amino acid 341 of SEQ ID NO:4, wherein said polypeptide is administered in an amount sufficient to reduce airway hyper responsiveness or reduce the increase of mucus-containing cells in the airway epithelium.

2. The method of claim 1 , wherein said polypeptide comprises from amino acid 26 to amino acid 341 of SEQ ID NO:4.

3. The method of claim 2 , wherein said polypeptide is part of fusion protein.

4. The method of claim 3 , wherein said fusion protein comprises an antibody fragment.

5. The method of claim 1 , wherein said polypeptide is part of a fusion protein.

6. The method of claim 5 , wherein said fusion protein comprises an antibody fragment.

7. The method of claim 6 , wherein said antibody fragment includes an Fc fragment.

8. The method of claim 1 , wherein the allergen-induced increase in airway hyper responsiveness is reduced.

9. The method of claim 1 , wherein the allergen-induced increase in mucus-containing cells in the airway epithelium is reduced.

10. A method of administering a polypeptide to a mammalian subject having allergen-induced airway hyper responsiveness, or allergen-induced increase of mucus-containing cells in an airway epithelium, said method comprising administering a polypeptide encoded by a polynucleotide that hybridizes under conditions comprising hybridization at 65° C. in 0.1×SSC to the complement of the portion of SEQ ID NO:3 that encodes from amino acid 26 to amino acid 341 of SEQ ID NO:4 in an amount sufficient to decrease airway hyper responsiveness or reduce the increase of mucus-containing cells in the airway epithelium, wherein said polypeptide is administered by oral ingestion, inhalation, or cutaneous, subcutaneous, or intravenous injection.

11. The method of claim 10 , wherein said polypeptide comprises from amino acid 26 to amino acid 341 of SEQ ID NO:4.

12. The method of claim 11 , wherein said polypeptide is part of a fusion protein.

13. The method of claim 10 , wherein said polypeptide is part of a fusion protein.

14. The method of claim 13 , wherein said fusion protein comprises an antibody fragment.

15. The method of claim 10 , wherein the subject has increased allergen-induced airway hyper responsiveness.

16. The method of claim 10 , wherein the subject has an increase in allergen-induced mucus-containing cells in the airway epithelium.

17. A method of treating an inflammatory condition of the lung characterized by an increase in allergen-induced airway hyper responsiveness and mucus-containing cells in a mammalian subject said method comprising administering to the subject a therapeutically effective amount of a polypeptide encoded by a polynucleotide that hybridizes under conditions comprising hybridization at 65° C. in 0.1×SSC to complement of the portion of SEQ ID NO:3 that encodes from amino acid 26 to amino acid 341 of SEQ ID NO:4 in an amount sufficient to decrease the lung inflammation in the subject.

18. The method of claim 17 , wherein said polypeptide comprises from amino acid 26 to amino acid 341 of SEQ ID NO:4.

19. The method of claim 18 , wherein said polypeptide is part of a fusion protein.

20. The method of claim 19 , wherein said fusion comprises an antibody fragment.

21. The method of claim 20 , wherein said antibody fragment includes an Fc fragment.

22. The method of any of claim 1 , 10 or 17 , wherein the administration of the polypeptide results in complete reversal of the allergen-induced airway hyper responsiveness.

23. The method of any of claim 1 , 10 or 17 , wherein the administration of the polypeptide results in complete reversal of the allergen-induced increase in mucus-containing cells in the airway epithelium.
