Abstract

The invention provides a novel GDP-mannose 4,6-dehydratase.

Claims (41)

1. A method for treating a subject having an inflammatory disorder, wherein the inflammatory disorder is characterized by excessive GDP-mannose 4,6 dehydratase (GM4,6D) activity or excessive GM4,6D nucleic acid expression, comprising administering a therapeutically effective amount of an antibody capable of binding to a polypeptide comprising the amino acid sequence of either SEQ ID NO:2 or SEQ ID NO: 3.

2. The method of claim 1 , wherein the antibody is a monoclonal antibody.

3. The method of claim 1 , wherein the antibody is a polyclonal antibody.

4. The method of claim 1 , wherein the antibody is capable of binding to a polypeptide comprising the amino acid sequence of SEQ ID NO: 3.

5. A method for treating a subject having an inflammatory disorder characterized by excessive GM4,6D activity or excessive GM4,6D nucleic acid expression and chosen frown arthritis, transplant rejection, asthma, sepsis, reperfusion injury, stroke and infection, comprising administering a therapeutically effective amount of an antibody capable of binding to a polypeptide comprising the amino acid sequence of either SEQ ID NO:2 or SEQ ID NO: 3.

6. The method of claim 5 , wherein the antibody is a monoclonal antibody.

7. The method of claim 5 , wherein the antibody is a polyclonal antibody.

8. The method of claim 5 , wherein the inflammatory disorder is arthritis.

9. The method of claim 5 , wherein the inflammatory disorder is transplant rejection.

10. The method of claim 5 , wherein the inflammatory disorder is asthma.

11. The method of claim 5 , wherein the inflammatory disorder is stroke.

12. The method of claim 5 , wherein the inflammatory disorder is infection.

13. A method of treating a subject having an inflammatory disorder characterized by excessive GM4,6D polypeptide activity comprising administering to the subject a compound, wherein said compound modulates GM4,6D polypeptide activity when combined with a substrate and a GM4,6D polypeptide chosen from:

(1) a polypeptide encoded by the nucleotide sequence of SEQ ID NO: 1;

(2) a polypeptide of SEQ ID NO: 2;

(3) a polypeptide of SEQ ID NO: 3;

(4) and a fragment of the polypeptide of SEQ ID NO:2 or SEQ ID NO: 3, which has GM4,6D activity.

14. The method of claim 13 , wherein the compound is an antibody that binds the GM4,6D polypeptide.

15. The method of claim 13 , wherein the antibody is a monoclonal antibody.

16. The method of claim 13 , wherein the antibody is a polyclonal antibody.

17. The method of claim 1 , wherein the antibody is capable of binding a polypeptide comprising the amino acid sequence of SEQ ID NO:2.

18. The method of claim 1 , wherein the antibody is capable of binding a polypeptide consisting of the amino acid sequence of SEQ ID NO:2.

19. The method of claim 1 , wherein the antibody is capable of binding a polypeptide consisting of the amino acid sequence of SEQ ID NO:3.

20. The method of claim 5 , wherein the antibody is capable of binding a polypeptide comprising the amino acid sequence of SEQ ID NO:2.

21. The method of claim 5 , wherein the antibody is capable of binding a polypeptide comprising the amino acid sequence of SEQ ID NO:3.

22. The method of claim 5 , wherein the antibody is capable of binding a polypeptide consisting of the amino acid sequence of SEQ ID NO:2.

23. The method of claim 5 , wherein the antibody is capable of binding a polypeptide consisting of the amino acid sequence of SEQ ID NO:3.

24. The method of claim 5 , wherein the inflammatory disorder is sepsis.

25. The method of claim 5 , wherein the inflammatory disorder is reperfusion injury.

26. The method of claim 14 , wherein the antibody is capable of binding a polypeptide comprising the amino acid sequence of SEQ ID NO:2.

27. The method of claim 14 , wherein the antibody is capable of binding a polypeptide comprising the amino acid sequence of SEQ ID NO:3.

28. The method of claim 14 , wherein the antibody is capable of binding a polypeptide consisting of the amino acid sequence of SEQ ID NO:2.

29. The method of claim 14 , wherein the antibody is capable of binding a polypeptide consisting of the amino acid sequence of SEQ ID NO:3.

30. The method of claim 13 , wherein the inflammatory disorder is chosen from arthritis, transplant rejection, asthma, sepsis, reperfusion injury, stroke and infection.

31. The method of 30 , wherein the inflammatory disorder is arthritis.

32. The method of claim 30 , wherein the inflammatory disorder is transplant rejection.

33. The method of claim 30 , wherein the inflammatory disorder is asthma.

34. The method of claim 30 , wherein the inflammatory disorder is sepsis.

35. The method of claim 30 , wherein the inflammatory disorder is reperfusion injury.

36. The method of claim 30 , wherein the inflammatory disorder is stroke.

37. The method of claim 30 , wherein the inflammatory disorder is infection.
