Abstract

The invention relates to α-galactosidase and to polynucleotides encoding the α-galactosidase. In addition methods of designing new α-galactosidases and method of use thereof are also provided. The α-galactosidases have increased activity and stability at increased pH and temperature.

Claims (11)

1. An isolated or recombinant polypeptide having at least 99% sequence identity to a polypeptide having a sequence as set forth in SEQ ID NO:4 and having α-galactosidase activity.

2. An isolated or recombinant polypeptide having at least 95% sequence identity to a polypeptide having a sequence as set forth in SEQ ID NO:4 as determined by analysis with a sequence comparison algorithm FASTA version 3.0t78 with the default parameters and having α-galactosidase activity.

3. An isolated or recombinant polypeptide having a sequence as set forth in SEQ ID NO:4.

4. An isolated or recombinant polypeptide, wherein the polypeptide is encoded by a polynucleotide having at least 95% sequence identity to SEQ ID NO:3, wherein the polypeptide has α-galactosidase activity.

5. An enzyme preparation comprising the polypeptide of claim 2 or claim 4 and a liquid, wherein the polypeptide has α-galactosidase activity.

6. A dry enzyme preparation comprising the polypeptide of claim 2 or claim 4 , wherein the polypeptide has α-galactosidase activity.

7. The isolated or recombinant polypeptide of claim 2 or claim 4 , wherein the α-galactosidase activity comprises the ability to renature and regain activity after exposure to temperatures of about 60° C. to about 105° C.

8. An isolated or recombinant polypeptide, wherein the polypeptide is encoded by a polynucleotide having at least 97% sequence identity to SEQ ID NO:3, wherein the polypeptide has α-galactosidase activity.

9. An isolated or recombinant polypeptide having α-galactosidase activity comprising an enzymatically active fragment of the polypeptide of claim 2 , claim 3 or claim 4 .

10. An enzyme preparation comprising the polypeptide of claim 3 and a liquid.

11. A dry enzyme preparation comprising the polypeptide of claim 3 .
