Method of inducing fetal hemoglobin synthesis
View Patent ↗The present invention is directed to secreted and transmembrane polypeptides and to nucleic acid molecules encoding those polypeptides. Also provided herein are vectors and host cells comprising those nucleic acid sequences, chimeric polypeptide molecules comprising the polypeptides of the present invention fused to heterologous polypeptide sequences, antibodies which bind to the polypeptides of the present invention and to methods for producing the polypeptides of the present invention.
1. A method comprising the steps of inducing a switch from adult hemoglobin to fetal hemoglobin in a cell comprising contacting said cell with a hemoglobin inducing polypeptide encoded by a nucleic acid comprising:
a) a nucleic acid sequence encoding the hemoglobin inducing polypeptide shown in FIG. 4 (SEQ ID NO:7);
b) a nucleic acid sequence encoding the hemoglobin inducing polypeptide shown in FIG. 4 (SEQ ID NO:7), lacking its associated signal peptide;
c) a nucleic acid shown in FIG. 3 (SEQ ID NO:6);
d) a full-length coding sequence of the nucleic acid sequence shown in FIG. 3 (SEQ ID NO:6); or
e) a full-length coding sequence of the cDNA deposited under ATCC accession number 209508.
2. The method of claim 1 , wherein the isolated nucleic acid comprises a nucleic acid sequence encoding the hemoglobin inducing polypeptide shown in FIG. 4 (SEQ ID NO:7).
3. The method of claim 1 , wherein the isolated nucleic acid comprises a nucleic acid sequence encoding the hemoglobin inducing polypeptide shown in FIG. 4 (SEQ ID NO:7), lacking its associated signal peptide.
4. The method of claim 1 , wherein the isolated nucleic acid comprises a nucleic acid shown in FIG. 3 (SEQ ID NO:6).
5. The method of claim 1 , wherein the isolated nucleic acid comprises a full-length coding sequence of the nucleic acid sequence shown in FIG. 3 (SEQ ID NO:6).
6. The method of claim 1 , wherein the isolated nucleic acid polypeptide comprises a full-length coding sequence of the cDNA deposited under ATCC accession number 209508.
7. A method comprising the steps of inducing a switch from adult hemoglobin to fetal hemoglobin in a cell comprising contacting said cell with a hemoglobin inducing polypeptide encoded by a nucleic acid comprising:
a) a nucleic acid sequence encoding the hemoglobin inducing polypeptide shown in FIG. 4 (SEQ ID NO:7) with 0-10 amino acid additions, deletions, or substitution;
b) a nucleic acid sequence encoding the hemoglobin inducing polypeptide shown in FIG. 4 (SEQ ID NO:7) with 0-10 amino acid additions, deletions, or substitutions, lacking its associated signal peptide;
c) a nucleic acid shown in FIG. 3 (SEQ ID NO:6);
d) a full-length coding sequence of the nucleic acid sequence shown in FIG. 3 (SEQ ID NO:6); or
e) a full-length coding sequence of the cDNA deposited under ATCC accession number 209508.
