Abstract

Compositions and methods are provided for the treatment and diagnosis of diseases amenable to modulation of the production of selected proteins. In accordance with preferred embodiments, oligonucleotides and oligonucleotide analogs are provided which are specifically hybridizable with a selected sequence of RNA or DNA wherein at least one of the 2′-deoxyfuranosyl moieties of the nucleoside unit is modified. Treatment of diseases caused by various viruses and other causative agents is provided.

Claims (6)

1. A mixed sequence oligonucleotide or oligonucleotide analog including more than one 2′-modified 2′-deoxyfuranosyl moiety wherein said modification comprises substitution by halo, azido, amino, alkoxy, thioalkoxy, alkylamino, or alkyl, and wherein one of said 2′-modified 2′-deoxyfuranosyl moieties is different from another of said 2′-modified 2′-deoxyfuranosyl moieties.

The mixed sequence oligonucleotide or oligonucleotide analog of claim 1 , wherein one of said 2′-modified 2′-deoxyfuranosyl moieties comprises substitution by halo, azido, amino, thioalkoxy, alkylamino or alkyl and another of said 2′-modified 2′-deoxyfuranosyl moieties comprises substitution by alkyloxy.

3. The mixed sequence oligonucleotide or oligonucleotide analog of claim 1 , wherein one of said 2′-modified 2′-deoxyfuranosyl moieties comprises substitution by halo, azido, amino, thioalkoxy or alkyl and another of said 2′-modified 2′-deoxyfuranosyl moieties comprises substitution by methoxy.

4. The mixed sequence oligonucleotide or oligonucleotide analog of claim 1 , wherein alkyl comprises a straight or branched chain alkyl.

5. The mixed sequence oligonucleotide or oligonucleotide analog of claim 4 , wherein alkyl includes unsaturation within the alkyl carbon chain.

6. The mixed sequence oligonucleotide of claim 1 wherein said oligonucleotide is a nuclease resistant oligonucleotide.
