Abstract

The present invention provides nucleic acids containing transcriptional units that encode CYP1B1 polypeptides or portions thereof, wherein the transcriptional units lack sequences found in the untranslated region (UTR) of naturally occurring forms of the CYP1B1 transcript. The nucleic acids of the invention lack translational repressor elements and thus provide for a system of enhanced translation of the CYP1B1 polypeptide or portions thereof. Also disclosed are methods of administering nucleic acids to a mammal and use in the treatment of proliferative disorders or cancer.

Claims (22)

1. A method of inducing an immune response in a mammal having a cancer, the method comprising administering to a mammal having a cancer an effective amount of a composition comprising a microparticle comprising a polymeric matrix or shell and a plasmid expression vector comprising a nucleic acid sequence encoding a polypeptide comprising SEQ ID NQ:38, wherein the nucleic acid sequence is operably linked to an expression control sequence, and wherein the composition is administered intramuscularly or subcutaneously.

2. The method of claim 1 , wherein the composition is administered intramuscularly.

3. The method of claim 1 , wherein the composition is administered subcutaneously.

4. The method of claim 1 , wherein the immune response is a T cell response.

5. The method of claim l, wherein the cancer is a cancer of the bladder, breast, colon, connective tissue, lung, esophagus, skin, lymph node, brain, ovary, stomach, uterus, testis, or prostate.

6. The method of claim 5 , wherein the cancer is a cancer of the colon.

7. The method of claim 5 , wherein the cancer is a cancer of the lymph node.

8. The method of claim 5 , wherein the cancer is a cancer of the ovary.

9. The method of claim 5 , wherein the cancer is a cancer of the prostate.

10. The method of claim 1 , wherein the polymeric matrix comprises poly lactide-co-glycolide (PLG).

11. The method of claim 1 , wherein the microparticle comprises a lipid.

12. The method of claim 10 , wherein the microparticle comprises a lipid.

13. A nucleic acid comprising a sequence encoding a polypeptide comprising SEQ ID NO:38.

14. A plasmid expression vector comprising a nucleic acid sequence encoding a polypeptide comprising SEQ ID NO:38, wherein the nucleic acid sequence is operably linked to an expression control sequence.

15. A microparticle comprising a polymeric matrix or shell and the nucleic acid of claim 13 .

16. The microparticle of claim 15 , wherein the microparticle comprises a polymeric matrix comprising PLG.

17. The microparticle of claim 15 , wherein the microparticle comprises a lipid.

18. The microparticle of claim 16 , wherein the microparticle comprises a lipid.

19. A microparticle comprising a polymeric matrix or shell and the expression vector of claim 14 .

20. The microparticle of claim 19 , wherein the microparticle comprises a polymeric matrix comprising PLG.

21. The microparticle of claim 19 , wherein the microparticle comprises a lipid.

22. The microparticle of claim 20 , wherein the microparticle comprises a lipid.
