Abstract

The invention relates to the genetic manipulation of plants to produce polyhydroxyalkanoate, particularly in the peroxisomes. Methods for producing such polymers in plants and host cells are provided. Such methods find use in producing biodegradable thermoplastics in plants and other organisms. Nucleotide molecules, expression cassettes, and genetically manipulated host cells, plants, plant tissues, and seeds are also provided.

Claims (32)

1. An isolated nucleotide molecule comprising a nucleotide sequence selected from the group consisting of:

(a) the nucleotide sequence set forth in SEQ ID NO: 1;

(b) a nucleotide sequence encoding the amino acid sequence set forth in SEQ ID NO: 2;

(c) a nucleotide sequence encoding a protein comprising 2-enoyl-CoA hydratase activity,

wherein said nucleotide sequence has at least 90% sequence identity to the nucleotide sequence set forth in SEQ ID NO: 1;

(d) the antisense nucleotide sequence complementary to the nucleotide sequence of (a), (b) or (c); and

(e) a nucleotide sequence encoding a protein comprising 2-enoyl-CoA hydratase activity,

wherein said nucleotide sequence hybridizes under high stringency conditions to the nucleotide sequence of (a); wherein high stringency conditions are hybridization in 50% formamide, 1M NaCl, 1% SDS at 37° C., and a wash in 0.1×SSC at 60 to 65° C.

2. An expression cassette comprising a nucleotide sequence of a nucleotide molecule of claim 1 , said nucleotide sequence operably linked to a promoter that drives expression in a host cell.

3. A host cell transformed with the expression cassette of claim 2 .

4. The host cell of claim 3 , wherein said host cell is selected from the group consisting of a plant cell, a bacterial cell, and a yeast cell.

5. The isolated nucleotide molecule of claim 1 , wherein said nucleotide sequence has at least 95% sequence identity to the nucleotide sequence set forth in SEQ ID NO; 1.

6. The isolated nucleotide molecule of claim 1 , wherein said nucleotide sequence is the nucleotide sequence set forth in SEQ ID NO: 1.

7. The isolated nucleotide molecule of claim 1 , wherein said nucleotide sequence encodes the amino acid sequence set forth in SEQ ID NO: 2.

8. A transgenic plant comprising in its genome at least one stably integrated DNA construct comprising a nucleotide sequence operably linked to a promoter that drives expression in a plant, wherein said nucleotide sequence is selected from the group consisting of:

(a) the nucleotide sequence set forth in SEQ ID NO: 1;

(b) a nucleotide sequence encoding the amino acid sequence set forth in SEQ ID NO: 2;

(c) a nucleotide sequence encoding a protein comprising 2-enoyl-CoA hydratase activity, wherein said nucleotide sequence has at least 90% sequence identity to the nucleotide sequence set forth in SEQ ID NO: 1;

(d) the antisense nucleotide sequence complementary to the nucleotide sequence of (a), (b) or (c); and

(e) a nucleotide sequence encoding a protein comprising 2-enoyl-CoA hydratase activity, wherein said nucleotide sequence hybridizes under high stringency conditions to the nucleotide sequence of (a): wherein high stringency conditions are hybridization in 50% formamide, 1M NaCl, 1% SDS at 37° C., and a wash in 0.1×SSC at 60 to 65°C.

9. The plant of claim 8 , wherein said promoters are selected from the group consisting of seed-preferred promoters, chemical-regulatable promoters, germination-preferred promoters, and leaf-preferred promoters.

10. The plant of claim 8 , wherein said plant is a monocot.

11. The plant of claim 10 , wherein said monocot is selected from the group consisting of maize, wheat, rice, sorghum, rye, millet, barley, palm and banana.

12. The plant of claim 8 , wherein said plant is a dicot.

13. The plant of claim 12 , wherein said dicot is selected from the group consisting of soybean, sunflower, safflower, alfalfa, potato, Brassica spp., cotton, tomato, tobacco and peanut.

14. A seed or the plant of claim 8 , wherein said seed comprises said DNA construct.

15. A transformed plant cell comprising in its genome at least one stably integrated DNA construct comprising a nucleotide sequence operably linked to a promoter that drives expression in a plant cell, wherein said nucleotide sequence is selected from the group consisting of:

(a) the nucleotide sequence set forth in SEQ ID NO: 1;

(b) a nucleotide sequence encoding the amino acid sequence set forth in SEQ ID NO: 2;

(c) a nucleotide sequence encoding a protein comprising 2-enoyl-CoA hydratase activity, wherein said nucleotide sequence has at least 90% sequence identity to the nucleotide sequence set forth in SEQ ID NO: 1;

(d) the antisense nucleotide sequence complementary to the nucleotide sequence of (a), (b) or (c); and

(e) a nucleotide sequence encoding a protein comprising 2-enoyl-CoA hydratase activity, wherein said nucleotide sequence hybridizes under high stringency conditions to the nucleotide sequence of (a); wherein high stringency conditions are hybridization in 50% formamide 1M NaCl, 1% SDS at 37° C., and a wash in 0.1×SSC at 60 to 65° C.
