Abstract

The invention provides compositions and methods for cell separation. These reagents and techniques specifically agglutinate cells via surface antigen recognition and can be used to recover even rare cell types in high yield.

Claims (40)

1. A regent comprising:

a) dextran;

b) anti-glycophorin A antibody;

c) anti-CD15 antibody; and

d) anti-CD9 antibody, wherein said anti-glycophorin A antibody, said anti-CD 15 antibody, and said anti-CD9 antibody are in solution.

2. The composition of claim 1 , further comprising anti-CD3 antibody.

3. The composition of claim 1 , further comprising anti-CD72 antibody.

4. The composition of claim 1 , further comprising anti-CD16 antibody.

5. The composition of claim 1 , further comprising anti-CD4 antibody.

6. The composition of claim 1 , further comprising anti-CD8 antibody.

7. The composition of claim 1 , further comprising anti-CD2 antibody.

8. The composition of claim 1 , further comprising heparin.

9. The composition of claim 1 , further comprising divalent cations.

10. The composition of claim 3 , wherein said divalent cations are Ca+2.

11. The composition of claim 3 , wherein said divalent cations are Mg+2.

12. The composition of claim 1 , further comprising phosphate buffered saline.

13. The composition of claim 1 , wherein the pH of said composition is between 6.8 to 7.8.

14. The composition of claim 1 , wherein the pH of said composition is between 7.2 to 7.4.

15. The composition of claim 1 , wherein said anti-glycophorin A antibody is monoclonal.

16. The composition of claim 1 , wherein said anti-glycophorin A antibody is an IgM antibody.

17. The composition of claim 1 , wherein said anti-glycophorin A antibody is an IgG antibody.

18. The composition of claim 1 , wherein said anti-glycophorin A antibody is an anti-human glycophorin A antibody.

19. The composition of claim 1 , wherein the concentration of said anti-glycophorin A antibody is about 0.1 mg/l to about 15 mg/l.

20. The composition of claim 1 , wherein said anti-CD15 antibody is a monoclonal antibody.

21. The composition of claim 1 , wherein said anti-CD15 antibody is an IgM antibody.

22. The composition of claim 1 , wherein said anti-CD15 antibody is an IgG antibody.

23. The composition of claim 1 , wherein said anti-CD15 antibody is an anti-human CD15 antibody.

24. The composition of claim 1 , wherein the concentration of said anti-CD15 antibody is about 0.1 mg/l to about 15 mg/l.

25. The composition of claim 1 , wherein said anti-CD9 antibody is a monoclonal antibody.

26. The composition of claim 1 , wherein said anti-CD9 antibody is an IgM antibody.

27. The composition of claim 1 , wherein said anti-CD9 antibody is an IgG antibody.

28. The composition of claim 1 , wherein said anti-CD9 antibody is an anti-human CD9 antibody.

29. The reagent of claim 1 , wherein the concentration of said anti-CD9 antibody is at about 0.1 mg/l to about 15 mg/l.

30. A reagent comprising:

a) dextran;

b) anti-glycophorin A antibody;

c) anti-CD15 antibody;

d) anti-CD9 antibody;

e) heparin; and

f) divalent cations, wherein said anti-glycophorin A antibody, said anti-CD15 antibody, and said anti-CD9 antibody are is solution.
