Compositions and methods for increasing bone mineralization
View Patent ↗Nucleic acids relating to human TGF-β binding proteins are disclosed. Also disclosed are assays for selecting molecules for increasing bone mineralization and methods for utilizing such molecules.
1. An isolated nucleic acid molecule comprising nucleotides 119 to 686 of the SEQ ID NO: 1 or the full complement thereof.
2. A vector comprising the nucleic acid molecule of claim 1 .
3. An isolated host cell transformed or transfected with the vector of claim 2 .
4. An isolated nucleic acid comprising the coding region of the nucleotide sequence of SEQ ID NO: 1 minus a secretion signal sequence, wherein said nucleic acid is obtainable by steps comprising:
(a) contacting a cDNA comprising the nucleotide sequence of SEQ ID NO: 1 with (i) the sense oligonucleotide primer comprising the sequence set forth in SEQ ID NO: 26 and (ii) the antisense oligonucleotide primer comprising the sequence set forth in SEQ ID NO: 27; and
(b) amplifying the cDNA under conditions and for a time sufficient to produce said nucleic acid molecule.
5. A vector comprising the nucleic acid according to claim 4 .
6. An isolated host cell that is transfected or transformed with the vector of claim 5 .
7. An isolated nucleic acid molecule comprising nucleotides 119 to 686 of the SEQ ID NO: 1.
8. A vector comprising the nucleic acid molecule of claim 7 .
9. An isolated host cell transformed or transfected with the vector of claim 8 .
10. A method for producing a polypeptide comprising the steps of:
(a) culturing the host cell according to claim 9 or 6 under conditions that permit expression of the polypeptide; and
(b) isolating said polypeptide.
11. An isolated polynucleotide encoding the secreted amino acid sequence of SEQ ID NO: 2.
12. An isolated host cell comprising a regulatory sequence operatively linked to a polynucleotide encoding the secreted amino acid sequence of SEQ ID NO: 2.
