Inhibition of tau-tau association
View Patent ↗A composition and use thereof for the prophylaxis or treatment of Alzheimer's disease, motor neurone disease, Lewy body disease, Pick's disease or progressive supranuclear palsy. The composition includes an agent which modulates or inhibits tau-tau association and which does not inhibit tau-tubulin binding. In the method the agent is administered to an individual in order to achieve the desired therapeutic effect. Preferably the agent which is administered is a phenothiazine compound.
1. A method for the treatment of Alzheimer's disease, motor neurone disease, Lewy body disease, Pick's disease, or progressive supranuclear palsy, which method comprises administering to a subiect in need thereof an effective amount of an agent which modulates or inhibits tau-tau association and which does not inhibit tau-tubulin binding.
2. A method according to claim 1 for the treatment of Alzheimer's disease.
3. A method according to claim 1 wherein said agent phenothiazines of the formulae:

R 1 , R 3 , R 4 , R 6 , R 7 and R 9 are independently selected from hydrogen, halogen, hydroxy, carboxy, substituted or unsubstituted alkyl, haloalkyl or alkoxy;
R 5 , each R 10 and each R 11 are independently selected from hydrogen, hydroxy, carboxy, substituted or unsubstituted alkyl, haloalkyl or alkoxy; and pharmaceutically acceptable salts thereof.
4. A method according to claim 3 wherein: R 1 , R 3 , R 4 , R 5 , R 6 , R 7 , R 9 , each R 10 and each R 11 are independently selected from hydrogen, —CH 3 , —C 2 H 5 or —C 3 H 7 .
5. A method according to claim 3 wherein said agent is a phenothiazine of the formula (I):
wherein:
R 1 , R 3 , R 4 , R 6 , R 7 and R 9 are independently selected from hydrogen, halogen, hydroxy, carboxy, substituted or unsubstituted alkyl, haloalkyl or alkoxy;
R 5 , R 10 and R 11 are independently selected from hydrogen, hydroxy, carboxy, substituted or unsubstituted alkyl, haloalkyl or alkoxy; and pharmaceutically acceptable salts thereof.
6. A method according to wherein R 5 is hydrogen.
7. A method according to claim 4 wherein said compound is selected from Methylene Blue, Toluidine Blue O, thionine, Azure A, Azure B or 1,9-dimethyl-Methylene Blue.
8. A method according to claim 3 wherein the agent is present in a therapeutically effective amount in a therapeutically inert carrier material in a pharmaceutical composition.
9. A method according to claim 3 wherein the agent has a standardized tau binding coefficient of less than 0.4 in the range 1-10 μg/ml, and does not inhibit tau-tubulin binding up to a molar ratio of 1000:1 with respect to the molar concentration of tau.
