Acellular dermal matrix and method of use thereof for grafting
View Patent ↗A method for processing and preserving an acellular collagen-based tissue matrix for transplantation is disclosed. The method includes the steps of processing biological tissues with a stabilizing solution to reduce procurement damage, treatment with a processing solution to remove cells, treatment with a cryoprotectant solution followed by freezing, drying, storage and rehydration under conditions that preclude functionally significant damage and reconstitution with viable cells.
1. An acellular tissue matrix made from dermal tissue comprising:
(a) an acellular reticular dermal matrix comprising collagen, wherein the acellular reticular dermal matrix comprises an extracellular protein and collagen matrix; and
(b) an intact basement membrane, wherein the basement membrane comprises a structurally preserved lamina densa and anchoring fibrils made by the process comprising:
de-epidermizing the dermal tissue with 1 M sodium chloride at 37±2° C. for 16-32 hours for human dermal tissue, or 35-55 hours for porcine dermal tissue,
removing the epidermis from the dermis,
rinsing the dermis with Hank's balanced salt solution at 20-26° C.,
de-cellularizing the dermis with 0.5% sodium dodecyl sulfate for 1 hour at 22-26° C., without the use of enzymatic treatment,
rinsing the dermis with Hank's balanced salt solution for 5 minutes at 20-26° C., thereby providing an acellular dermal matrix,
wherein the acellular dermal matrix is not subjected to a cross-linking agent.
2. The acellular dermal matrix of claim 1 , wherein the dermal tissue is human.
3. The acellular dermal matrix of claim 1 , wherein the dermal tissue is porcine.
4. The acellular dermal matrix of claim 1 , wherein the acellular dermal matrix has been freeze-dried.
5. The acellular dermal matrix of claim 1 , wherein the acellular dermal matrix is capable of supporting neovascularization when implanted in or on a patient.
6. The acellular dermal matrix of claim 1 , wherein the acellular dermal matrix is capable of supporting fibroblast infiltration when implanted in or on a patient.
7. The acellular dermal matrix of claim 1 , wherein the acellular dermal matrix is capable of supporting growth of keratinocytes.
8. The acellular dermal matrix of claim 1 , wherein the basement membrane allow attachment of epithelial and endothelial cells.
9. A method of treatment comprising grafting the acellular dermal matrix of claim 1 to or into a patient.
10. The method of claim 9 , wherein the patient is selected from the group consisting of a burn patient, a diabetic patient, a patient with pressure ulcers, a patient requiring reconstructive surgery, and a patient requiring skin replacement following excision of a skin lesion.
