Abstract

The present invention relates to compounds according to the general formula (I) which bind to the NR1H4 receptor and act as agonists of the NR1H4 receptor. The invention further relates to the treatment of diseases and/or conditions through binding of the nuclear receptor by the compounds and the production of medicaments using the compounds.

Claims (40)

1. A compound including resolved diastereoisomers and enantiomers, and tautomers, pharmaceutical acceptable salts or solvates thereof, having the following formula (I):

wherein:

R 1 is selected from the group consisting of C 1 to C 7 substituted alkyl, substituted phenyl, C 7 to C 12 phenylalkyl, C 7 to C 12 substituted phenylalkyl, C 2 to C 7 substituted heterocyclic ring, C 5 to C 6 heteroaryl, and C 5 to C 6 substituted heteroaryl;

R 2 is selected from the group consisting of C 1 to C 7 substituted alkyl, substituted phenyl, C 7 to C 12 phenylalkyl, C 7 to C 12 substituted phenylalkyl, C 2 to C 7 substituted heterocyclic ring, C 5 to C 6 heteroaryl, and C 5 to C 6 substituted heteroaryl;

R 3 is hydrogen;

R 4 is selected from the group consisting of C 1 to C 8 substituted alkyl,

wherein the alkyl is substituted with one or more (substituted or unsubstituted) C 3 to C 7 cycloalkyl, C 7 to C 12 substituted phenylalkyl,

wherein the phenyl is substituted with one or more substituents selected from the group consisting of halogen, protected hydroxy, cyano, nitro, C 2 to C 8 alkyl, C 1 to C 8 substituted alkyl, C 1 to C 8 alkoxy, C 1 to C 8 substituted alkoxy, C 1 to C 8 acyl, C 1 to C 8 substituted acyl, to C 1 to C 8 acyloxy, carboxy, protected carboxy, carboxymethyl, protected carboxymethyl, hydroxymethyl, protected hydroxymethyl, protected amino, (monosubstituted)amino, protected (monosubstitued)amino, (disubstituted)amino, carboxyamide, N,N-di(C 1 to C 6 alkyl)carboxamide, trifluoromethyl, N—((C 1 to C 6 alkyl)sulfonyl)amino, N-(phenylsulfonyl)amino, cyclic C 2 to C 8 alkylene and a phenyl group, substituted or unsubstituted and substituted naphthyl.

2. The compound of claim 1 , wherein:

R 1 is selected from the group consisting of C 1 to C 7 substituted alkyl, substituted phenyl, C 7 to C 12 phenylalkyl, C 7 to C 12 substituted phenylalkyl, C 2 to C 7 substituted heterocyclic ring, C 5 to C 6 heteroaryl, and C 5 to C 6 substituted heteroaryl;

R 2 is selected from the group consisting of C 1 to C 7 substituted alkyl, phenyl, substituted phenyl, C 7 to C 12 phenylalkyl, C 7 to C 12 substituted phenylalkyl, C 7 to C 12 substituted phenylalkyl, C 2 to C 7 substituted heterocyclic ring, C 5 to C 6 heteroaryl, and C 5 to C 6 substituted heteroaryl;

R 3 is hydrogen;

R 4 is:

3. The compound of claim 1 , wherein:

R 1 is selected from the group consisting of C 1 to C 7 substituted alkyl, substituted phenyl, C 7 to C 12 phenylalkyl, C 7 to C 12 substituted phenylalkyl, C 2 to C 7 substituted heterocyclic ring, C 5 to C 6 heteroaryl, and C 5 to C 6 substituted heteroaryl;

R 2 is selected from the group consisting of C 1 to C 7 substituted alkyl, substituted phenyl, C 7 to C 12 phenylalkyl, C 7 to C 12 substituted phenylalkyl, C 2 to C 7 substituted heterocyclic ring, C 5 to C 6 heteroaryl and C 5 to C 6 substituted heteroaryl;

R 3 is hydrogen; and

R 4 is:

18. A therapeutic composition comprising at least one compound according to any one of claims 1 to 17 in admixture with a pharmaceutically acceptable carrier, adjuvant or vehicle.

19. A compound including resolved diastereoisomers and enantiomers, and tautomers, pharmaceutical acceptable salts or solvates thereof, having the following formula (I):

R 1 is selected from the group consisting of C 1 to C 7 substituted alkyl, substituted phenyl, C 7 to C 12 phenylalkyl, C 7 to C 12 substituted phenylalkyl, C 2 to C 7 substituted heterocyclic ring, C 5 to C 6 heteroaryl, and C 5 to C 6 substituted heteroaryl;

R 2 is selected from the group consisting of C 1 to C 7 substituted alkyl, substituted phenyl, C 7 to C 12 phenylalkyl, C 7 to C 12 substituted phenylalkyl, C 2 to C 7 substituted heterocyclic ring, C 5 to C 6 heteroaryl, and C 5 to C 6 substituted heteroaryl;

R 3 is hydrogen;

R 4 is selected from the group consisting of the following structures:

wherein n is an integer from 1 to 3.
