Abstract

The present invention provides an adsorbent chip, which includes three components, a substrate, an intermediate layer of linker arms and an adsorbent film, which is attached to the linker arms. The adsorbent film is made up of a plurality of adsorbent particles, each of which includes a binding functionality. The invention also provides a method of making the chips of the invention in which the substrate-intermediate film cassette is formed and the adsorbent film is subsequently immobilized thereon. When the adsorbent film is from the same preparation across a particular batch of chips, the chips provide for the acquisition of data that are highly reproducible from one chip to the next throughout the particular batch of chips. Additionally, the invention provides methods for using the chips to perform assays.

Claims (87)

1. A method of detecting an analyte, said method comprising:

(a) contacting said analyte with a chip comprising:

(i) a planar substrate comprising a surface;

(ii) an intermediate layer covalently attached to said surface, wherein said intermediate layer comprises charged linker arms; and

(iii) an adsorbent film attached to said intermediate layer by an electrostatic interaction, said adsorbent film comprising a plurality of charged latex particles bound to said linker arms wherein each said charged latex particle has a diameter of about 3 microns to about 500 microns, and wherein each said charged latex particle comprises a binding functionality

wherein said binding functionality interacts with said analyte, thereby adsorbing said analyte on said adsorbent film, and

wherein said charged linker arms and said charged latex particles are oppositely charged; and

(b) desorbing said analyte and detecting said desorbed analyte using mass spectrometry.

2. The method according to claim 1 , wherein said surface comprises at least one addressable feature, said addressable feature having said intermediate layer attached thereto and said intermediate layer having said adsorbent film attached thereto.

3. The method according to claim 1 , wherein said binding functionality is selected from the group consisting of positively charged functinctionalities, negatively charged functionalities, metal ion functionalities, polar functionalities, hydrophobic functionalities and combinations thereof.

4. The method according to claim 1 , wherein said binding functionality comprises a biomolecule.

5. The method according to claim 1 , wherein said particles are from about 3 microns to about 10 microns in diameter.

6. The method according to claim 1 , wherein said particles are from about 10 microns to about 500 microns in diameter.

7. The method according to claim 1 , wherein the analyte is detected by laser desorption/ionization mass spectrometry.

8. The method according to claim 7 , said method further comprising:

(c) prior to step (b), washing said chip, thereby removing analyte not interacting with a binding functionality.

9. The method according to claim 1 , said chip comprising:

(i) an aluminum substrate comprising a silicon dioxide surface;

(ii) an intermediate layer attached to said surface, said intermediate layer comprising a moiety having the formula:

wherein n is an integer greater than 1; and

(iii) an adsorbent film immobilized electrostatically on said intermediate layer, said adsorbent film comprising a plurality of adsorbent particles comprising the subunit:

10. The method according to claim 1 , said chip comprising:

(i) said substrate is an aluminum substrate comprising a silicon dioxide surface;

(ii) said intermediate layer comprises a polymer formed by co-polymerization of acrylic acid and a propylmethacrylate moiety immobilized on said silicon dioxide surface through a Si—O—Si bond; and

(iii) said adsorbent film comprises a plurality of charged latex particles comprising the subunit:

11. The method according to claim 10 , said intermediate layer comprising a moiety having the formula:

12. The method according to claim 1 , said chip comprising:

(i) an aluminum substrate comprising a silicon dioxide surface;

(ii) an intermediate layer attached to said surface, said intermediate layer comprising a polymer formed by co-polymerization of N-(3-N,N-dimethylaminopropyl)methacrylamide and a propylmethacrylate moiety immobilized on said surface through a Si—O—Si bond; and

(iii) an adsorbent film immobilized electrostatically on said intermediate layer, said adsorbent film comprising a plurality of adsorbent particles comprising the subunit:

13. The method according to claim 12 , said intermediate layer comprising a moiety having the formula:

14. The method according to claim 1 , wherein

(i) said substrate is an aluminum substrate comprises a silicon dioxide surface;

(ii) said intermediate layer comprises a moiety having the formula:

(iii) said adsorbent particles comprise the subunit:

15. The method according to claim 1 , wherein

(i) said substrate is an aluminum substrate comprising a silicon dioxide surface;

(ii) said intermediate layer comprises a moiety having the formula:

and

(iii) said adsorbent particles comprise the subunit:

16. The method according to claim 1 , wherein

(i) said substrate is an aluminum substrate comprising a silicon dioxide surface;

(ii) said intermediate layer comprises a moiety having the formula:

and

(iii) said adsorbent particles comprise the subunit:

17. A method of detecting an analyte, said method comprising

(a) contacting said analyte with a chip comprising:

(i) an aluminum substrate comprising a silicon dioxide surface;

(ii) an intermediate layer comprising a moiety having the formula:

wherein

n is an integer greater than 1; and

(iii) an adsorbent film attached to said intermediate layer by an electrostatic interaction, said adsorbent film comprising a plurality of charged latex particles comprising the subunit:

wherein each said latex particle as a diameter from about 3 microns to about 500 microns; and

(b) desorbing said analyte and detecting said desorbed analyte using mass spectrometry.

18. A method of detecting an analyte, said method comprising:

(a) contacting said analyte with a chip comprising:

(i) a planar substrate comprising a surface;

(ii) an intermediate layer covalently attached to said surface, wherein said intermediate layer comprises charged linker arms; and

(iii) an adsorbent film attached to said intermediate layer by an electrostatic interaction, said adsorbent film comprising a plurality of charged latex particles bound to said linker arms,

wherein said charged latex particles comprise a subunit which is a member selected from:

and

wherein each said charged latex particle has a diameter of from about 3 microns to about 500 microns,

wherein each said charged latex particle comprises a binding functionality

wherein said binding functionality interacts with said analyte, thereby adsorbing said analyte on said adsorbent film, and

wherein said charged linker arms and said charged latex particles are oppositely charged; and

(b) desorbing said analyte and detecting said desorbed analyte using mass spectrometry.

19. A method of detecting an analyte, said method comprising:

(a) contacting said analyte with a chip comprising:

(i) an aluminum substrate comprising a silicon dioxide surface;

(ii) an intermediate layer covalently attached to said surface, wherein said intermediate layer comprises charged linker arms; and

(iii) an adsorbent film attached to said intermediate layer by an electrostatic interaction, said adsorbent film comprising a plurality of charged latex particles bound to said linker arms,

wherein each said charged particle has a diameter of from about 3 microns to about 500 microns,

wherein each said charged latex particle comprises a binding functionality

wherein said binding functionality interacts with said analyte, thereby adsorbing said analyte on said adsorbent film, and

wherein said charged linker arms and said charged latex particles are oppositely charged; and

(b) desorbing said analyte and detecting said desorbed analyte using mass spectrometry.
