Curcuminoid compositions exhibiting synergistic inhibition of the expression and/or activity of cyclooxygenase-2
View Patent ↗Compositions comprising an effective amount of a curcuminoid species and an effective amount of a diterpene lactone species, a triterpene species or derivatives thereof that have a synergistic effect on specific inhibition of inducible COX-2 activity and have minimal effect on COX-1 activity are disclosed. Methods of using the compositions for providing synergistic anti-inflammatory effects are also disclosed.
1. A composition comprising an effective amount of curcumin and an effective amount of andrographolide, wherein the ratio of curcumin and andrographolide is from 1:10 to 10:1, said composition having at least 5 times less inhibitory activity on cyclooxygenase-1 (COX-1) than on cyclooxygenase-2 (COX-2).
2. The composition of claim 1 , wherein the curcumin is pharmaceutical grade curcumin.
3. The composition of claim 1 , wherein the andrographolide is pharmaceutical grade andrographolide.
4. The composition of claim 1 , wherein at least one of the curcumin or andrographolide is derived from a plant or plant extract.
5. The composition of claim 1 , wherein at least one of the curcumin or andrographolide is conjugated with a compound selected from the group consisting of mono- or disaccharides, amino acids, sulfates, succinate, acetate and glutathione.
6. The composition of claim 5 , wherein the conjugated compound is mono- or disaccharide.
7. The composition of claim 1 , said composition further comprising one or more members selected from the group consisting of antioxidants, vitamins, minerals, proteins, fats, carbohydrates, glucosamine, chondrotin sulfate and aminosugars.
8. The composition of claim 7 , wherein said composition further comprises glucosamine.
9. A composition comprising an effective amount of curcumin and an effective amount of oleanolic acid, wherein the ratio of curcumin and oleanolic acid is from 1:10 to 10:1, said composition having at least 5 times less inhibitory activity on cyclooxygenase-1 (COX-1) than on cyclooxygenase-2 (COX-2).
10. The composition of claim 9 , wherein the curcumin is pharmaceutical grade curcumin.
11. The composition of claim 9 , wherein the oleanolic acid is pharmaceutical grade oleanolic acid.
12. The composition of claim 9 , wherein at least one of the curcumin or oleanolic acid is derived from a plant or plant extract.
13. The composition of claim 9 , wherein at least one of the curcumin or oleanolic acid is conjugated with a compound selected from the group consisting of mono- or di-saccharides, amino acids, sulfates, succinate, acetate and glutathione.
14. The composition of claim 13 , wherein the conjugated compound is mono- or di-saccharide.
15. The composition of claim 9 , said composition further comprising one or more members selected from the group consisting of antioxidants, vitamins, minerals, proteins, fats, carbohydrates, glucosamine, chondrotin sulfate and aminosugars.
16. The composition of claim 15 , wherein said composition further comprises glucosamine.
