Abstract

The invention discloses methods of proliferation and differentiation of multipotent neural stem cells. Also provided are methods of making cDNA libraries and methods of screening biological agents which affect proliferation differentiation survival phenotype or function of CNS cells.

Claims (12)

1. A method of proliferating a culture of undifferentiated neural cells containing one or more multipotent neural stem cells, wherein said undifferentiated neural cells are nestin + and are glial fibrillary acidic protein (GFAP) − ; neurofilament (NF) − ; neuron specific enolase (NSE) − ; and myelin basic protein (MBP) − , the method comprising culturing the undifferentiated neural cells in a culture medium containing at least one proliferation-inducing growth factor.

2. The method of claim 1 , wherein the proliferation-inducing growth factor is selected from the group consisting of epidermal growth factor, amphiregulin, acidic fibroblast growth factor, basic fibroblast growth factor, transforming growth factor alpha, and combinations thereof.

3. The method of claim 1 , wherein said culture medium is serum-free.

4. The method of claim 1 , wherein the undifferentiated neural cells are human cells.

5. The method of claim 1 , wherein the undifferentiated neural cells are cultured in a suspension culture.

6. The method of claim 1 , wherein the undifferentiated neural cells are cultured in an adherent culture.

A method of proliferating a culture of undifferentiated neural cells containing one or more multipotent neural stem cells, wherein said undifferentiated neural cells stain positive for nestin and said undifferentiated neural cells lack differentiated neural cells that do not stain positive for nestin and that stain positive for the differentiated neural cell markers neurofilament, glial fibrillary acidic protein, neuron specific enolase, and myclin basic protein, the method comprising culturing the undifferentiated neural cells in a culture medium containing at least one proliferation-inducing growth factor.

8. The method of claim 7 , wherein the proliferation-inducing growth factor is selected from the group consisting of epidermal growth factor, amphiregulin, acidic fibroblast growth factor, basic fibroblast growth factor, transforming growth factor alpha, and combinations thereof.

9. The method of claim 7 , wherein said culture medium is serum-free.

10. The method of claim 7 , wherein the undifferentiated neural cells are human cells.

11. The method of claim 7 , wherein the undifferentiated neural cells are cultured in a suspension culture.

12. The method of claim 7 , wherein the undifferentiated neural cells are cultured in an adherent culture.
