Abstract

The invention provides novel LSR genomic sequences, polypeptides, antibodies, and polynucleotides including biallelic markers derived from the LSR locus. Primers hybridizing to regions flanking these biallelic markers are also provided. This invention also provides polynucleotides and methods suitable for genotyping a nucleic acid containing sample for one or more biallelic markers of the invention. Further, the invention provides methods to detect a statistical correlation between a biallelic marker allele and a phenotype and/or between a biallelic marker haplotype and a phenotype.

Claims (25)

1. A method for determining whether an individual is at risk of developing a detectable trait or whether a detectable trait that an individual suffers from is statistically associated with one or more LSR biallelic markers, comprising:

obtaining a nucleic acid sample from said individual; and

determining the identity of the polymorphic base of said one or more LSR biallelic marker in said nucleic acid sample;

wherein the detectable trait is obesity or an obesity-related metabolic disorder; and wherein the LSR biallelic marker is selected from SNP #1 (A15; position 3778 of SEQ ID NO: 1), SNP #2 (A17; position 15007 of SEQ ID NO: 1), SNP #3 (A21; position 19744 of SEQ ID NO: 1) or SNP #4 (A30; position 359 of SEQ ID NO: 12).

2. A method of using an anti-obesity drug, comprising:

obtaining a nucleic acid sample from an individual;

determining the identity of a polymorphic base of one or more LSR biallelic markers selected from SNP #1 (A15; position 3778 of SEQ ID NO: 1), SNP #2 (A17; position 15007 of SEQ ID NO: 1), SNP #3 (A21; position 19744 of SEQ ID NO: 1) or SNP #4 (A30; position 359 of SEQ ID NO: 12) in said nucleic acid sample; and

administering said drug to said individual if the genotype of SNP #1 is CT or TT and/or SNP #2 is AA and/or SNP #3 is GG.

3. A method of screening an individual for inclusion in a clinical trial of an anti-obesity drug comprising:

obtaining a nucleic acid sample from said individual;

determining the identity of the polymorphic base of one or more LSR biallelic markers selected from SNP #1 (A15; position 3778 of SEQ ID NO: 1), SNP #2 (A17; position 15007 of SEQ ID NO: 1), SNP #3 (A21; position 19744 of SEQ ID NO: 1) or SNP #4 (A30; position 359 of SEQ ID NO: 12) in said nucleic acid sample; and

including said individual in said clinical trial if the genotype of SNP #1 is CT or TT and/or SNP #2 is AA and/or SNP #3 is GG.

4. The method of claim 1 , wherein said detectable trait is obesity.

5. The method of claim 1 , wherein said obesity-related metabolic disorder is type II diabetes.

6. The method of claim 4 , wherein obesity is characterized by plasma triglyceride levels above the mean of the population.

7. The method of claim 4 , wherein obesity is characterized by total cholesterol levels above the mean of the population.

8. The method of claim 4 , wherein obesity is characterized by free fatty acid levels above the mean of the population.

9. The method of claim 4 , wherein obesity is characterized by an insulin level relative to body mass index (BMI) above the mean of the population.

10. The method of claim 5 , wherein said obesity-related disorder is characterized by an level of glucose tolerance above the mean of the population.

11. The method of claim 4 , wherein the genotype at SNP #1 is CT or TT.

12. The method of claim 4 , wherein the genotype at SNP #2 is AA.

13. The method of claim 4 , wherein the genotype of SNP #3 is GG.

14. The method of claim 5 , wherein the genotype at SNP #1 is CT or TT.

15. The method of claim 5 , wherein the genotype at SNP #2 is AA.

16. The method of claim 5 , wherein the genotype of SNP #3 is GG.
