Abstract

The invention provides methods and compositions relating to an IκB kinase, IKK-α, and related nucleic acids. The polypeptides may be produced recombinantly from transformed host cells from the disclosed IKK-α encoding nucleic acids or purified from human cells. The invention provides isolated IKK-α hybridization probes and primers capable of specifically hybridizing with the disclosed IKK-α genes, IKK-α-specific binding agents such as specific antibodies, and methods of making and using the subject compositions in diagnosis, therapy and in the biopharmaceutical industry.

Claims (20)

1. An isolated or recombinant polynucleotide encoding a polypeptide comprising at least 10 consecutive residues of the amino acid sequence set forth as SEQ ID NO:4, which consecutive amino acid residues comprise (a) at least one of the amino acid residues 679, 680, 684, 686 and 687 of SEQ ID NO:4 and (b) the amino acid residue 543 (Glu) of SEQ ID NO:4.

An isolated or recombinant polynucleotide according to claim 1 , wherein said polypeptide has an activity selected from at least one of: a kinase or kinase inhibitory activity, a NIK-binding or binding inhibitory activity, an IκB-binding or binding inhibitory activity and an NFκB activating or inhibitory activity.

3. An isolated or recombinant polynucleotide according to claim 1 comprising at least 24 consecutive nucleotides of the nucleotide sequence set forth as SEQ ID NO:3, which consecutive nucleotides comprise nucleotide 1628 (A) of SEQ ID NO:3.

4. An isolated or recombinant polynucleotide according to claim 3 , comprising at least 36 consecutive nucleotides of the nucleotide sequence set forth as SEQ ID NO:3, which consecutive nucleotides comprise the nucleotide 1628 (A) of SEQ ID NO:3.

5. An isolated or recombinant polynucleotide according to claim 3 , comprising at least 96 consecutive nucleotides of the nucleotide sequence set forth as SEQ ID NO:3, which consecutive nucleotides comprise the nucleotide 1628 (A) of SEQ ID NO:3.

6. An isolated or recombinant polynucleotide according to claim 3 , wherein the consecutive nucleotides comprise nucleotides 1611–1628 of SEQ ID NO:3.

7. An isolated or recombinant polynucleotide according to claim 3 , wherein the consecutive nucleotides comprise nucleotides 1628–1644 of SEQ ID NO:3.

8. An isolated or recombinant polynucleotide according to claim 3 , wherein the consecutive nucleotides comprise nucleotides 1620–1648 of SEQ ID NO:3.

9. An isolated or recombinant polynucleotide according to claim 3 , wherein the consecutive nucleotides comprise nucleotides 1612–1648 of SEQ ID NO:3.

10. An isolated or recombinant polynucleotide according to claim 3 , wherein the consecutive nucleotides comprise nucleotides 1608–1668 of SEQ ID NO:3.

11. An isolated or recombinant polynucleotide according to claim 3 , wherein the consecutive nucleotides comprise nucleotides 1580–1650 of SEQ ID NO:3.

12. An isolated or recombinant polynucleotide according to claim 3 , wherein the consecutive nucleotides comprise nucleotides 1596–1636 of SEQ ID NO:3.

13. An isolated or recombinant polynucleotide according to claim 3 , wherein the consecutive nucleotides comprise nucleotides 1602–1687 of SEQ ID NO:3.

14. An isolated or recombinant polynucleotide according to claim 3 , wherein the consecutive nucleotides comprise nucleotides 1614–1639 of SEQ ID NO:3.

15. An isolated or recombinant polynucleotide according to claim 3 , wherein the consecutive nucleotides comprise nucleotides 1621–1753 of SEQ ID NO:3.

16. An isolated or recombinant polynucleotide according to claim 3 , wherein the consecutive nucleotides comprise nucleotides 1502–1899 of SEQ ID NO:3.

17. An isolated or recombinant polynucleotide according to claim 3 , wherein the consecutive nucleotides comprise nucleotides 1020–1790 of SEQ ID NO:3.

18. A method of making a polypeptide, said method comprising steps: introducing a polynucleotide according to claim 1 into a host cell or cellular extract, incubating said host cell or extract under conditions whereby said nucleic acid is expressed as a transcript and said transcript is expressed as a translation product comprising said polypeptide.

19. A cell comprising a polynucleotide according to claim 1 .

20. A cell comprising a polynucleotide according to claim 3 .
