Abstract

A filter assembly for separating and collecting a layer of particulate matter (e.g., cells) from a liquid containing the particulate matter (e.g., a biological fluid specimen). The filter assembly has a holder and contiguous inner and outer fluid pervious media. The outer fluid pervious medium is attached to the sidewall of the holder. The inner fluid pervious medium is of low fluidic impedance relative to the outer fluid pervious medium. The filter assembly is designed for use in a separation housing that defines two flow paths between inlet and outlet, one directly through the filter and the other around the periphery of the filter. The inlet portion of the housing has a central inlet in a radially sloped surface that faces the filter collection site. A movable suction head is adapted to cooperate with the housing and the filter assembly.

Claims (33)

1. An apparatus, for use with a filter, for separating and collecting a layer of particulate matter from a liquid containing the particulate matter, the filter having a collection site adapted to collect a layer of the particulate matter, the apparatus comprising a particulate matter separation chamber for holding the filter, the separation chamber defined at least in part by a bottom wall with a central fluid inlet and an annular wall projecting upwardly from the bottom wall, the bottom wall having a plurality of circumferentially spaced projections adjacent the annular wall that contact the filter adjacent its periphery, when installed in the separation chamber with the collection site facing the bottom wall, to maintain a gap between the filter and the bottom wall for outflow of liquid, the bottom wall further having a radially sloped surface facing the filter and configured such that the distance between the bottom wall and the collection site decreases from the inlet toward the filter periphery, whereby liquid introduced to the separation chamber through the inlet can flow directly through the filter and deposit a layer of particulate matter on the collection site, and can also flow with particulate matter outwardly across the face of the filter and through the spaces at the gap between the projections.

2. An apparatus according to claim 1 , wherein the spaces between the projections are large enough to prevent the particulate matter from clogging the spaces.

3. An apparatus according to claim 2 , adapted for collecting a layer of cells taken from a cytology specimen, wherein the minimum dimension of the spaces between the projections is 0.004 in.

4. An apparatus according to claim 1 , wherein the sloped surface of the bottom wall is substantially conical.

An apparatus according to claim 1 , wherein the inlet flow area, which is defined by the product of the circumference of the inlet and the distance between the inlet and the collection site, is substantially equal to the aggregate of the radial outflow areas of the spaces between the projections.

An apparatus according to any one of claims 1 to 5 , for use in a container for holding and processing particulate matter-containing liquid and adapted to be engaged by an external manipulator while in the container, further comprising a tube communicating with the separation chamber through the inlet and extending downwardly from the bottom wall, whereby the tube can contact the particulate matter containing-liquid in the container.

7. An apparatus according to claim 6 , further comprising a dispersing element carried by the tube for mixing the particulate matter containing-liquid in the container.

8. An apparatus according to claim 6 , wherein the external manipulator is a vacuum head for applying suction to the separation chamber to draw particulate matter-containing liquid through the filter, and the separation chamber is adapted to mate with and seal to the vacuum head.

9. An apparatus for separating and collecting a layer of particulate matter from a liquid containing the particulate matter, comprising:

a housing defining a separation chamber having an inlet portion and an outlet portion; and

a filter in the separation chamber having a collection site facing the inlet portion and defining two flow paths between the inlet portion and the outlet portion, one flow path directly through the filter and the other flow path around the periphery of the filter,

wherein the inlet portion has a central inlet in a surface that faces the collection site and is radially sloped such that the distance between the sloped surface and the collection site decreases from the inlet toward the periphery of the filter,

and wherein the housing comprises a plurality of circumferentially spaced filter-contacting projections adjacent the periphery of the filter that maintain a gap for peripheral outflow of liquid between the filter and the inlet portion,

whereby liquid introduced to the separation chamber through the inlet flows along said one flow path directly through the filter to deposit a layer of particulate matter on the collection site, and also flows with particulate matter along said other flow path outwardly across the collection site, through the spaces at the gap between the projections and around the periphery of the filter to wash excess particulate matter from the collection site.

10. An apparatus according to claim 9 , wherein the sloped surface is substantially radially symmetrical.

11. An apparatus according to claim 9 , wherein the inlet flow area, which is defined by the product of the circumference of the inlet and the distance between the inlet and the collection site, is substantially equal to the aggregate radial outflow area at the periphery of the filter.

A method for collecting cells for cytology comprising passing a biological fluid containing cells through a separation chamber having therein a filter with a cell collection site positioned transversely of the fluid flow path entering the separation chamber through a centrally located inlet, and diverting a portion of the fluid flow outwardly toward the periphery of the filter, and through a plurality of discrete flow passages adjacent the periphery that communicate with a common outlet downstream of the filter, by juxtaposition of an inlet surface adjacent the collection site that is radially sloped such that the distance between the sloped surface and the collection site decreases from the inlet toward the periphery of the filter, whereby fluid introduced to the separation chamber through the inlet flows directly through the filter to deposit cells on the collection site, and also flows outwardly with particulate matter across the collection site toward the periphery of the filter to wash away excess cells from the collection site leaving substantially a monolayer of cells on the collection site.

13. A method according to claim 12 , wherein the inlet flow area, which is defined by the product of the circumference of the inlet and the distance between the inlet and the collection site, is equal to or greater than the aggregate radial outflow area at the periphery of the filter.

A method according to claim 12 , wherein the collection site is slightly bowed outwardly at its center, further comprising removing the filter from the separation chamber and pressing the filter against a slide to flatten it against the slide and transfer substantially all of the cells from the filter to the slide.

15. An apparatus according to claim 1 , wherein the bottom wall of the separation chamber is devoid of filter-contacting portions in the central region thereof bounded by the circumferentially spaced projections.

16. An apparatus according to claim 1 , wherein the radially sloped surface of the bottom wall is devoid of protrusions.

17. An apparatus according to claim 16 , wherein the radially sloped surface of the bottom wall is substantially smooth.

18. An apparatus according to claim 9 , wherein the radially sloped surface is devoid of filter-contacting portions.

19. An apparatus according to claim 9 , wherein the radially sloped surface is devoid of protrusions.

20. An apparatus according to claim 19 , wherein the radially sloped surface is substantially smooth.

21. A method according to claim 12 , wherein the radially sloped surface is devoid of filter-contacting portions.

22. A method according to claim 12 , wherein the radially sloped surface is devoid of protrusions.

23. A method according to claim 22 , wherein the radially sloped surface is substantially smooth.

24. An apparatus, for use with a filter, for separating and collecting a layer of particulate matter from a liquid containing the particulate matter, the filter having a collection site adapted to collect a layer of the particulate matter, the apparatus comprising a particulate matter separation chamber for holding the filter, the separation chamber defined at least in part by a bottom wall with a central fluid inlet and an annular wall projecting upwardly from the bottom wall, the bottom wall having a plurality of circumferentially spaced projections adjacent the annular wall that contact the filter adjacent its periphery, when installed in the separation chamber with the collection site facing the bottom wall, to maintain a gap between the filter and the bottom wall for outflow of liquid, the bottom wall further having a radially sloped surface facing the filter and configured such that the distance between the bottom wall and the collection site continuously decreases from the inlet to the projections, the radially sloped surface being devoid of filter-contacting portions, whereby liquid introduced to the separation chamber through the inlet can flow directly through the filter and deposit a layer of particulate matter on the collection site, and can also flow with particulate matter outwardly across the face of the filter and through the spaces at the gap between the projections.

25. An apparatus according to claim 24 , wherein the radially sloped surface is devoid of protrusions.

26. An apparatus according to claim 25 , wherein the radially sloped surface is substantially smooth.

27. An apparatus according to claim 26 , wherein the radially sloped surface is substantially conical.

28. An apparatus according to claim 24 , wherein the radially sloped surface is substantially conical.
