Abstract

A method is presented for killing a first population of cells within a mixture of viable cells by providing the mixture of viable cells, contacting the cells with a label, illuminating a portion of the mixture, capturing an image of multiple cells in the illuminated portion of the mixture, determining at least two dimensional coordinates of one or more cells of the first population of cells using the first captured image, and applying a lethal dose of energy to said dimensional coordinates of the one or more cells at a first focal plane.

Claims (40)

1. A method of killing a first population of cells within a mixture of cells, comprising:

a) providing a mixture of cells comprising viable cells;

b) contacting said mixture of cells with a label to yield a first population of cells that is labeled and second population of cells that is not labeled, such that the first population can be distinguished from the second population;

c) illuminating a portion of said mixture of cells;

d) capturing a first image of a plurality of cells in the illuminated portion of said mixture of cells;

e) determining at least two dimensional coordinates of one or more cells of said first population of cells by reference to the first captured image and said label; and

f) applying a lethal dose of energy to said dimensional coordinates of said one or more cells at a first focal plane;

wherein one or more cells of said second population of cells remains viable.

2. The method of claim 1 , wherein said lethal dose of energy irreversibly damages cellular components of said cell, leading to cell death without destruction of the cellular membrane.

3. The method of claim 1 , wherein said mixture of cells is dispersed in a sub-confluent monolayer.

4. The method of claim 1 , further comprising:

g) applying another lethal dose of energy to said dimensional coordinates of said one or more cells at a second focal plane.

5. The method of claim 1 , further comprising:

g) capturing a second image of said plurality of cells in said illuminated portion of said mixture of cells;

h) determining at least two dimensional coordinates of one or more remaining viable cells of said first population of cells by reference to the second captured image and said label; and

i) applying a lethal dose of energy to said dimensional coordinates of said one or more remaining viable cells at said first focal plane.

6. The method of claim 5 , wherein said lethal dose of energy irreversibly damages one or more cellular components of said cell, leading to cell death without destruction of the cellular membrane.

7. The method of claim 5 , wherein said mixture of cells is dispersed in a sub-confluent monolayer before the capturing a first image step is performed.

8. The method of claim 5 , further comprising:

j) applying a lethal dose of energy to said dimensional coordinates of said one or more remaining viable cells at a second focal plane.

9. A method of killing a first population of cells within a mixture of cells, said method comprising:

a) providing a mixture of cells comprising viable cells;

b) contacting said mixture of cells with a label to yield a first population of cells that is not labeled and a second population of cells that is labeled, such that the first population can be distinguished from the second population;

c) illuminating a portion of said mixture of cells;

d) capturing a first image of a plurality of cells in the illuminated portion of said mixture of cells;

e) determining at least two dimensional coordinates of one or more cells of said first population by reference to the first captured image; and

f) applying a lethal dose of energy to said dimensional coordinates of said one or more cells at a first focal plane;

wherein one or more cells of said second population of cells remains viable.

10. The method of claim 9 , wherein said lethal dose of energy irreversibly damages cellular components of said cell, leading to cell death without destruction of the cellular membranes.

11. The method of claim 9 , wherein said mixture of cells is dispersed in a sub-confluent monolayer.

12. The method of claim 9 , further comprising:

g) applying another lethal dose of energy to said dimensional coordinates of said one or more cells at a second focal plane.

13. The method of claim 9 , further comprising:

g) capturing a second image of said plurality of cells in said illuminated portion of said mixture of cells;

h) determining at least two dimensional coordinates of one or more remaining viable cells of said first population of cells by reference to the second captured image; and

i) applying another lethal dose of energy to said dimensional coordinates of said one or more cells at said first focal plane.

14. The method of claim 13 , wherein said lethal dose of energy irreversibly damages cellular components of said cell, leading to cell death without destruction of the cellular membranes.

15. The method of claim 13 , wherein said mixture of cells is dispersed in a sub-confluent monolayer before the capturing a first image step is performed.

16. The method of claim 13 , further comprising:

j) applying another lethal dose of energy to said dimensional coordinates of said one or more cells at a second focal plane.
