Abstract

The invention relates to reverse focusing and methods and systems which employ reverse focusing. In a particular example, an automated focusing microscope uses one or more reverse focusing steps to acquire a focused image. Computer programs having instructions for instructing a computer to acquire a focused image by reverse focusing are also included.

Claims (6)

1. A method for automatic focusing of an image of a sample on a slide comprising:

(a) positioning a focal plane of an objective at a first z-position between the bottom surface of the slide and the top surface of the slide and acquiring a first image;

(b) reverse focusing along a z-axis at a plurality of z-positions;

(c) acquiring a plurality of images at each of the plurality of z-positions;

(d) processing the images in order to identify a peak focus power of each image; and

(e) selecting the peak focus power to obtain a focused image of the sample positioned on the slide.
