Abstract

The nucleotide and polypeptide sequences of two novel human flavin-containing monooxygenase (FMO) enzymes are presented: hFMO2 and hFMOx. Vectors and host cells for cloning and/or expression of hFMO2 and hFMOx are described, as are methods of preparing hFMO2 and hFMOx polypeptides. Also included are methods for selecting compounds, diagnosing predisposition to pathologies and/or deficiencies related to FMO's, and pharmaceutical compositions containing compounds for treating and/or preventing these pathologies.

Claims (57)

1. An isolated, purified, or recombinant polynucleotide:

a) encoding a polypeptide comprising the amino acid sequence of SEQ ID NO: 3, wherein said polypeptide has flavin monooxygenase activity;

b) encoding a polypeptide comprising SEQ ID NO: 3;

c) encoding a polypeptide comprising a fragment of SEQ ID NO: 3, wherein said polypeptide and said fragment has flavin monooxygenase activity;

d) encoding a polypeptide consisting of a fragment of SEQ ID NO: 3; or

e) comprising a polynucleotide sequence completely complementary to a polynucleotide as set forth in a), b), c), or d).

2. The isolated, purified, or recombinant polynucleotide according to claim 1 , wherein the polynucleotide encodes a polypeptide comprising the amino acid sequence of SEQ ID NO: 3, wherein said polypeptide has flavin monooxygenase activity.

3. The isolated, purified, or recombinant polynucleotide according to claim 1 , wherein the polynucleotide encodes a polypeptide comprising a fragment of SEQ ID NO: 3, wherein said polypeptide and said fragment has flavin monooxygenase activity.

4. The isolated, purified, or recombinant polynucleotide according to claim 1 , wherein said polynucleotide encodes a polypeptide comprising SEQ ID NO: 3.

5. The isolated, purified, or recombinant polynucleotide according to claim 1 , wherein said polynucleotide encodes a polypeptide consisting of a fragment of SEQ ID NO: 3.

The isolated, purified, or recombinant polynucleotide according to claim 1 , wherein the polynucleotide comprises a polynucleotide sequence completely complementary to an isolated, purified, or recombinant polynucleotide encoding a polypeptide comprising the amino acid sequence of SEQ ID NO: 3, wherein said polypeptide has flavin monooxygenase activity.

7. The isolated, purified, or recombinant polynucleotide according to claim 1 , wherein the polynucleotide comprises a polynucleotide sequence completely complementary to an isolated, purified, or recombinant polynucleotide encoding a polypeptide comprising SEQ ID NO: 3.

The isolated, purified, or recombinant polynucleotide according to claim 1 , wherein the polynucleotide comprises a polynucleotide sequence completely complementary to an isolated, purified, or recombinant polynucleotide encoding a polypeptide comprising a fragment of SEQ ID NO: 3, wherein said polypeptide and said fragment has flavin monooxygenase activity.

9. The isolated, purified, or recombinant polynucleotide according to claim 1 , wherein the polynucleotide comprises a polynucleotide sequence completely complementary to an isolated, purified, or recombinant polynucleotide encoding a polypeptide consisting of a fragment of SEQ ID NO: 3.

10. A cloning or expression vector comprising an isolated, purified, or recombinant polynucleotide:

a) encoding a polypeptide comprising the amino acid sequence of SEQ ID NO: 3, wherein said polypeptide has flavin monooxygenase activity;

b) encoding a polypeptide comprising SEQ ID NO: 3;

c) encoding a polypeptide comprising a fragment of SEQ ID NO: 3, wherein said polypeptide and said fragment has flavin monooxygenase activity;

d) encoding a polypeptide consisting of a fragment of SEQ ID NO: 3; or

e) comprising a polynucleotide sequence completely complementary to a polynucleotide as set forth in a), b), c), or d).

11. The cloning or expression vector according to claim 10 , wherein the vector is an expression vector.

12. The cloning or expression vector according to claim 10 , wherein the vector is a cloning vector.

13. The cloning or expression vector according to claim 10 , wherein the vector further comprises promoter sequences and regulatory sequences.

14. The cloning or expression vector according to claim 11 , wherein the vector further comprises promoter sequences and regulatory sequences.

15. The cloning or expression vector according to claim 12 , wherein the vector further comprises promoter sequences and regulatory sequences.

16. The cloning or expression vector according to claim 10 , wherein the vector comprises a polynucleotide encoding a polypeptide comprising the amino acid sequence of SEQ ID NO: 3, wherein said polypeptide has flavin monooxygenase activity.

17. The cloning or expression vector according to claim 10 , wherein the vector comprises a polynucleotide encoding a polypeptide comprising a fragment of SEQ ID NO: 3, wherein said polypeptide and said fragment has flavin monooxygenase activity.

18. The cloning or expression vector according to claim 10 , wherein the vector comprises a polynucleotide encoding a polypeptide comprising SEQ ID NO: 3.

19. The cloning or expression vector according to claim 10 , wherein the vector comprises a polynucleotide encoding a polypeptide consisting of a fragment of SEQ ID NO: 3.

The cloning or expression vector according to claim 10 , wherein the vector comprises a polynucleotide comprising a polynucleotide sequence completely complementary to an isolated, purified, or recombinant polynucleotide encoding a polypeptide comprising the amino acid sequence of SEQ ID NO: 3, wherein said polypeptide has flavin monooxygenase activity.

21. The cloning or expression vector according to claim 10 , wherein the vector comprises a polynucleotide comprising a polynucleotide sequence completely complementary to an isolated, purified, or recombinant polynucleotide encoding a polypeptide comprising SEQ ID NO: 3.

The cloning or expression vector according to claim 10 , wherein the vector comprises a polynucleotide comprising a polynucleotide sequence completely complementary to an isolated, purified, or recombinant polynucleotide encoding a polypeptide comprising a fragment of SEQ ID NO: 3, wherein said polypeptide and said fragment has flavin monooxygenase activity.

23. The cloning or expression vector according to claim 10 , wherein the vector comprises a polynucleotide comprising a polynucleotide sequence completely complementary to an isolated, purified, or recombinant polynucleotide encoding a polypeptide consisting of a fragment of SEQ ID NO: 3.

The cloning or expression vector according to claim 10 , wherein said vector comprises a polynucleotide sequence completely complementary to an isolated, purified, or recombinant polynucleotide encoding a polypeptide comprising the amino acid sequence of SEQ ID NO: 3, wherein said polypeptide has flavin monooxygenase activity.

25. A host cell comprising a polynucleotide or vector according to claim 2 , 3 , 4 , 5 , 6 , 7 , 8 , 9 , 11 , 12 , 13 , 14 , 15 , 16 , 17 , 18 , 19 , 20 , 21 , 22 , 23 , or 24 .

26. The host cell according to claim 25 , wherein said host cell comprises a polynucleotide according to claim 2 .

27. The host cell according to claim 25 , wherein said host cell comprises a polynucleotide according to claim 3 .

28. The host cell according to claim 25 , wherein said host cell comprises a polynucleotide according to claim 4 .

29. The host cell according to claim 25 , wherein said host cell comprises a polynucleotide according to claim 5 .

30. The host cell according to claim 25 , wherein said host cell comprises a polynucleotide according to claim 6 .

31. The host cell according to claim 25 , wherein said host cell comprises a polynucleotide according to claim 7 .

32. The host cell according to claim 25 , wherein said host cell comprises a polynucleotide according to claim 8 .

33. The host cell according to claim 25 , wherein said host cell comprises a polynucleotide according to claim 9 .

34. The host cell according to claim 25 , wherein said host cell comprises a vector according to claim 11 .

35. The host cell according to claim 25 , wherein said host cell comprises a vector according to claim 12 .

36. The host cell according to claim 25 , wherein said host cell comprises a vector according to claim 13 .

37. The host cell according to claim 25 , wherein said host cell comprises a vector according to claim 14 .

38. The host cell according to claim 25 , wherein said host cell comprises a vector according to claim 15 .
