Abstract

Isolatable, hydroxyapatite-targeting polymeric structures, and biologically active conjugates thereof, are provided. The polymeric structure includes a linear or branched water-soluble and non-peptidic polymer backbone, such as a PEG backbone, having at least two termini, a first terminus being covalently bonded to a hydroxyapatite-targeting moiety, such as a bisphosphonate, and a second terminus covalently bonded to a chemically reactive group, wherein said chemically reactive group is protected or unprotected. Methods of preparing and using hydroxyapatite-targeting polymeric structures, and biologically active conjugates thereof, are also provided.

Claims (23)

1. A method for preparing a bioactive agent hydroxyapatite-targeting polymer conjugate, said method comprising the steps of:

a. providing an isolated water-soluble polymer comprising, at a first terminus, a hydroxyapatite-targeting moiety, and at a second terminus, a reactive group suitable for reacting with a functional group of a biologically active agent, and

b. reacting said isolated water-soluble polymer from step (a) with a biologically active agent comprising said functional group under conditions effective to form a covalently bonded biologically active agent-water-soluble polymer-hydroxyapatite targeting conjugate,

wherein said biologically active agent comprises multiple functional groups suitable for reacting with said reactive group, and said reacting step is carried out under conditions effective to form a mixture of biologically active agent-water-soluble polymer-hydroxyapatite targeting conjugates, wherein said mixture comprises different conjugate multimer species each having a different number of hydroxyapatite-targeting-water-soluble polymers covalently attached to said biologically active agent, wherein said mixture is purified to separate each of the conjugate multimer species.

2. The method of claim 1 , wherein said method further comprises, prior to said providing step, purifying said isolated water-soluble polymer from step (a).

3. The method of claim 2 , wherein said purifying step comprises precipitation followed by solvent extraction, or chromatography.

4. The method of claim 3 , wherein said purifying step comprises ion exchange chromatography.

5. The method of claim 1 , wherein said conjugate is effective to bind to hydroxyapatite in vitro when incubated therewith for a period of at least an hour and a half under conditions effective to promote hydroxyapatite-binding.

6. The method of claim 1 , wherein said biologically active agent is a peptide or protein.

7. The method of claim 1 , wherein said biologically active agent comprises at least 3 functional groups capable of reacting with said reactive groups.

8. The method of claim 7 , wherein said mixture comprises different conjugate species having 1-, 2-, or 3- hydroxyapatite-targeting water soluble polymers covalently attached to said biologically active agent.

9. The method of claim 1 , wherein said hydroxyapatite-targeting moiety is selected from the group consisting of tetracycline, calcein, bisphosphonates, polyaspartic acid, polyglutamic acid, and aminophosphosugars.

10. The method of claim 9 , wherein said hydroxyapatite-targeting moiety is a bisphosphonate.

11. The method of claim 10 , wherein said bisphosphonate is selected from the group consisting of etidronate, alendronate, and 3-amino-1-hydroxypropane-1-1-diphosphonic acid.

The method of claim 1 , wherein said water-soluble polymer is selected from the group consisting of poly(alkylene glycol), poly(oxyethylated polyol), poly(olefinic alcohol), poly(vinylpyrolidone), poly(hydroxypropylmethacrylamide), poly(α-hydroxy acid), poly(vinyl alcohol), polyphosphazene, polyoxazoline, and copolymers and terpolymers thereof.

13. The method of claim 12 , wherein said water-soluble polymer is poly(ethylene glycol).

14. The method of claim 1 , wherein said water-soluble polymer is linear or branched.

15. The method of claim 1 , wherein said water-soluble polymer has a molecular weight ranging from about 100 daltons to about 100,000 daltons.

16. The method of claim 15 , wherein said water-soluble polymer has a molecular weight ranging from about 6,000 daltons to about 80,000 daltons.

17. The method of claim 1 , wherein said biologically active agent is selected from the group consisting of peptides, proteins, small molecule drugs, dyes, nucleosides, oligonucleotides, lipids, phospholipids, cells, viruses, liposomes, microparticles and micelles.

18. The method of claim 17 , wherein said biologically active agent is selected from the group consisting of peptides, proteins, and small molecule drugs.

The method of claim 1 , wherein said reactive group is selected from the group consisting of hydroxyl, ester, carbonate, acetal, aldehyde, aldehyde hydrate, alkenyl, acrylate, methacrylate, acrylamide, sulfone, amine, hydrazide, thiol, carboxylic acid, isocyanate, isothiocyanate, maleimide, vinylsulfone, dithiopyridine, vinylpyridine, iodoacetamide, epoxide, glyoxal, dione, mesylate, tosylate, and tresylate.

20. The method of claim 19 , wherein said reactive group is selected from the group consisting of hydroxyl, amine, carboxylic acid, maleimide, carbonate and ester.
