Abstract

Polymeric derivatives of indoles such as are disclosed. Methods of making and using the same as well as related polymeric conjugates are also disclosed.

Claims (120)

1. A compound of the formula:

wherein:

R 1 is a polyalkylene oxide;

Y 1 is O, S or NR 2 ;

R 2 is selected from the group consisting of hydrogen, C 1-6 alkyls, C 3-12 branched alkyls, C 3-8 cycloalkyls, C 1-6 substituted alkyls, C 3-8 substituted cyloalkyls, aryls, substituted aryls, aralkyls, C 1-6 heteroalkyls, substituted C 1-6 heteroalkyls, C 1-6 alkoxy, phenoxy and C 1-6 heteroalkoxy;

L 1 is a bifunctional linker selected from the group consisting of

—NH(CH 2 CH 2 O) n (CH 2 ) n NR 3 —,

—NH(CH 2 CH 2 O) n C(O)—,

—NH(CR 4 R 5 ) n OC(O)—,

—C(O)(CR 4 R 5 ) n NHC(O)(CR 8 R 7 ) q NR 3 —,

—C(O)O(CH 2 ) n O—,

—C(O)(CR 4 R 5 ) n NR 3 —,

—C(O)NH(CH 2 CH 2 O) n (CH 2 ) n NR 3 —,

—C(O)O − (CH 2 CH 2 O) n NR 3 —,

—C(O)NH(CR 4 R 5 ) n O—,

—C(O)O(CR 4 R 5 ) n O—,

—C(O)NH(CH 2 CH 2 O) n —,

wherein

R 3 , R 4 , R 5 , R 7 , and R 8 are independently selected from the group consisting of hydrogen, C 1-6 alkyls, C 3-12 branched alkyls, C 3-8 cycloalkyls, C 1-6 substituted alkyls, C 3-8 substituted cyloalkyls, aryls substituted aryls, aralkyls, C 1-6 heteroalkyls, substituted C 1-6 heteroalkyls, C 1-6 alkoxy, phenoxy and C 1-6 heteroalkoxy;

R 6 is selected from the group consisting of hydrogen, C 1-6 alkyls, C 3-12 branched alkyls, C 3-8 cycloalkyls, C 1-6 substituted alkyls, C 3-8 substituted cyloalkyls, aryls substituted aryls, aralkyls, C 1-6 heteroalkyls, substituted C 1-6 heteroalkyls, C 1-6 alkoxy, phenoxy and C 1-6 heteroalkoxy, NO 2 , haloalkyl and halogen; and

n and q are each a positive integer;

p is 0 or 1; and

B is

wherein

Y 7 is O, S or NR 12′ ; and

R 1′ -R 12′ are each independently selected from the group consisting of hydrogen, C 1-6 alkyls, C 3-12 branched alkyls, C 3-8 cycloalkyls, C 1-6 substituted alkyls, C 3-12 alkenyls, C 3-12 substituted alkenyls, C 3-12 alkynyls, C 3-12 substituted alkynyls, C 3-8 substituted cycloalkyls, aryls, substituted aryls, aralkyls, C 1-6 heteroalkyls, substituted C 1-6 heteroalkyls, C 1-6 alkoxy, phenoxy, C 1-6 heteroalkoxy, halo-, nitro-, cyano-, hydroxyl-, amino-, carboxy- and trifluoromethyl,

(i) wherein the substituted alkyls, substituted cycloalkyls, substituted aryls and substituted heteroalkyls are independently substituted with the group consisting of halo, alkoxy, nitro, carboxy, amino, alkylamino, hydroxyl and mercapto; and

(ii) wherein a carbon of the heteroalkyls and heteroalkoxy is independently substituted with the group consisting of O, N and S.

2. A compound of claim 1 , wherein Y 1 is O.

3. The compound of claim 1 wherein R 2 is selected from the group consisting of hydrogen, methyl and ethyl.

4. The compound of claim 3 wherein R 2 is hydrogen.

5. The compound of claim 1 wherein said paullone is a member of the group consisting of alsterpaullone and kenpaullone.

6. The compound of claim 1 , wherein R 1 is a polyethylene glycol.

7. The compound of claim 6 , wherein R 1 is selected from the group consisting of

—O—C(O)CH 2 —O—(CH 2 CH 2 O x —CH 2 C(O)—)—,

—NR 3 CH 2 CH 2 —O—(CH 2 CH 2 O) x —CH 2 CH 2 NR 3 —,

—SHCH 2 CH 2 —O—(CH 2 CH 2 O) x —CH 2 CH 2 SH—,

wherein

x is the degree of polymerization; and

R 3 is selected from the group consisting of hydrogen, C 1-6 alkyls, C 3-12 branched alkyls, C 3-8 cycloalkyls, C 1-6 substituted alkyls, C 3-8 substituted cycloalkyls, aryls substituted aryls, aralkyls, C 1-6 heteroalkyls, substituted C 1-6 heteroalkyls, C 1-6 alkoxy, phenoxy and C 1-6 heteroalkoxy,

(i) wherein the substituted alkyls, substituted cycloalkyls, substituted aryls and substituted heteroalkyls are independently substituted with the group consisting of halo, alkoxy, nitro, carboxy, amino, alkylamino, hydroxyl and mercapto; and

(ii) wherein a carbon of the heteroalkyls and heteroalkoxy is independently substituted with the group consisting of O, N and S.

8. The compound of claim 1 , wherein R 1 is —O—(CH 2 CH 2 O) x — and x is a positive integer so that the weight average molecular weight is at least about 20,000 Da.

9. The compound of claim 1 , wherein R 1 has a weight average molecular weight of from about 20,000 Da to about 100,000 Da.

10. The compound of claim 1 , wherein R 1 has a weight average molecular weight of from about 25,000 Da to about 60,000 Da.

11. A compound of claim 1 , selected from the group consisting of:

R 4 , R 5 and R 6 are independently selected from the group consisting of is selected from the group consisting of hydrogen, C 1-6 alkyls, C 3-12 branched alkyls, C 3-8 cycloalkyls, C 1-6 substituted alkyls, C 3-8 substituted cycloalkyls, aryls substituted aryls, aralkyls, C 1-6 heteroalkyls, substituted C 1-6 heteroalkyls, C 1-6 alkoxy, phenoxy and C 1-6 heteroalkoxy;

B is

Y 2 is O, S or NR 12′ ;

R 1′ -R 12′ are each independently selected from the group consisting of hydrogen, C 1-6 alkyls, C 3-12 branched alkyls, C 3-8 cycloalkyls, C 1-6 substituted alkyls, C 3-12 alkenyls, C 3-12 substituted alkenyls, C 3-12 alkynyls, C 3-12 substituted alkynyls, C 3-8 substituted cycloalkyls, aryls, substituted aryls, aralkyls, C 1-6 heteroalkyls, substituted C 1-6 heteroalkyls, C 1-6 alkoxy, phenoxy, C 1-6 heteroalkoxy, halo-, nitro-, cyano-, hydroxyl-, amino-, carboxy- and trifluormethyl,

(i) wherein the substituted alkyls, substituted cycloalkyls, substituted aryls and substituted heteroalkyls are independently substituted with the group consisting of halo, alkoxy, nitro, carboxy, amino, alkylamino, hydroxyl and mercapto; and

(ii) wherein a carbon of the heteroalkyls and heteroalkoxy is independently substituted with the group consisting of O, N and S.

12. A compound of claim 11 wherein said paullone is a member of the group consisting of alsterpaullone and kenpaullone.

13. A method of preparing a polymer conjugate of the formula:

R 1 is a polyalkylene oxide;

L 1 is a bifunctional spacer selected from the group consisting of

—NH(CH 2 CH 2 O) n (CH 2 ) n NR 3 —,

—NH(CH 2 CH 2 O) n C(O)—,

—NH(CR 4 R 5 ) n NHC(O)(CR 8 R 7 ) q NR 3 —,

—C(O)(CR 4 R 5 ) n NHC(O)(CR 8 R 7 ) n NR 3 —,

—C(O)O(CH 2 ) n O—,

—C(O)(CR 4 R 5 ) n NR 3 —,

—C(O)NH(CH 2 CH 2 O) n (CH 2 ) n NR 3 —,

—C(O)O − (CH 2 CH 2 O) n NR 3 —,

—C(O)NH(CR 4 R 5 ) n O—,

—C(O)O(CR 4 R 5 ) n O—,

—C(O)NH(CH 2 CH 2 O) n —,

wherein

R 3 , R 4 , R 5 , R 7 , and R 8 are independently selected from the group consisting of hydrogen, C 1-6 alkyls, C 3-12 branched alkyls, C 3-8 cycloalkyls, C 1-6 substituted alkyls, C 3-8 substituted cycloalkyls, aryls substituted aryls, aralkyls, C 1-6 heteroalkyls, substituted C 1-6 heteroalkyls, C 1-6 alkoxy, phenoxy and C 1-6 heteroalkoxy;

R 6 is selected from the group consisting of hydrogen, C 1-6 alkyls, C 3-12 branched alkyls, C 3-8 cycloalkyls, C 1-6 substituted alkyls, C 3-8 substituted cycloalkyls, aryls substituted aryls, aralkyls, C 1-6 heteroalkyls, substituted C 1-6 heteroalkyls, C 1-6 alkoxy, phenoxy and C 1-6 heteroalkoxy, NO 2 , haloalkyl and halogen; and

n and q are each a positive integer;

Y 1 is O, S or NR 2 ;

R 2 is selected from the group consisting of hydrogen, C 1-6 alkyls, C 3-12 branched alkyls, C 3-8 cycloalkyls, C 1-6 substituted alkyls, C 3-8 substituted cycloalkyls, aryls substituted aryls, aralkyls, C 1-6 heteroalkyls, substituted C 1-6 heteroalkyls, C 1-6 alkoxy, phenoxy and C 1-6 heteroalkoxy;

p is 0 or 1; and

B is

wherein

Y 2 is O, S or NR 12 ; and

R 1 , -R 12 , are each independently selected from the group consisting of hydrogen, C 1-6 alkyls, C 3-12 branched alkyls, C 3-8 cycloalkyls, C 1-6 substituted alkyls, C 3-12 alkenyls, C 3-12 substituted alkenyls, C 3-12 alkynyls, C 3-12 substituted alkynyls, C 3-8 substituted cycloalkyls, aryls, substituted aryls, aralkyls, C 1-6 heteroalkyls, substituted C 1-6 heteroalkyls, C 1-6 alkoxy, phenoxy, C 1-6 heteroalkoxy, halo-, nitro-, cyano-, hydroxyl-, amino-, carboxy- and trifluormethyl,

(i) wherein the substituted alkyls, substituted cycloalkyls, substituted aryls and substituted heteroalkyls are independently substituted with the group consisting of halo, alkoxy, nitro, carboxy, amino, alkylamino, hydroxyl and mercapto; and

(ii) wherein a carbon of the heteroalkyls and heteroalkoxy is independently substituted with the group consisting of O, N and S;

comprising:

a) acylating a paullone,

b) attaching a blocked bifunctional spacer to form a blocked intermediate, and

c) deblocking said blocked intermediate and reacting the deblocked intermediate with an activated polymer

under conditions sufficient to cause a polymeric conjugate to be formed.

14. A method of preparing a polymer conjugate of the formula:

R 1 is a polyalkylene oxide;

L 1 is a bifunctional spacer selected from the group consisting of

—NH(CH 2 CH 2 O) n (CH 2 ) n NR 3 —,

—NH(CH 2 CH 2 O) n C(O)—, —NH(CR 4 R 5 ) n OC(O)—,

—C(O)(CR 4 R 5 ) n NHC(O)(CR 8 R 7 ) q NR 3 —,

—C(O)O(CH 2 ) n O—,

—C(O)(CR 4 R 5 ) n NR 3 —,

—C(O)NH(CH 2 CH 2 O) n (CH 2 ) n NR 3 —,

—C(O)O − (CH 2 CH 2 O) n NR 3 —,

—C(O)NH(CR 4 R 5 ) n )—,

—C(O)NH(CH 2 CH 2 O) n —,

wherein

R 3 , R 4 , R 5 , R 7 , and R 8 are independently selected from the group consisting of hydrogen, C 1-6 alkyls, C 3-12 branched alkyls, C 3-8 cycloalkyls, C 1-6 substituted alkyls, C 3-8 substituted cyloalkyls, aryls substituted aryls, aralkyls, C 1-6 heteroalkyls, substituted C 1-6 heteroalkyls, C 1-6 alkoxy, phenoxy and C 1-6 heteroalkoxy;

R 6 is selected from the group consisting of hydrogen, C 1-6 alkyls, C 3-12 branched alkyls, C 3-8 cycloalkyls, C 1-6 substituted alkyls, C 3-8 substituted cycloalkyls, aryls substituted aryls, aralkyls, C 1-6 heteroalkyls, substituted C 1-6 heteroalkyls, C 1-6 alkoxy, phenoxy and C 1-6 heteroalkoxy, NO 2 , haloalkyl and halogen; and

n and q are each a positive integer;

Y 1 is O, S or NR 2 ;

R 2 is selected from the group consisting of hydrogen, C 1-6 alkyls, C 3-12 branched alkyls, C 3-8 cycloalkyls, C 1-6 substituted alkyls, C 3-8 substituted cycloalkyls, aryls substituted aryls, aralkyls, C 1-6 heteroalkyls, substituted C 1-6 heteroalkyls, C 1-6 alkoxy, phenoxy and C 1-6 heteroalkoxy;

p is 0 or 1; and

B is

wherein

Y 2 is O, S or NR 12′ ; and

R 1 ,-R 12 , are each independently selected from the group consisting of hydrogen, C 1-6 alkyls, C 3-12 branched alkyls, C 3-8 cycloalkyls, C 1-6 substituted alkyls, C 3-12 alkenyls, C 3-12 substituted alkenyls, C 3-12 alkynyls, C 3-12 substituted alkynyls, C 3-8 substituted cycloalkyls, aryls, substituted aryls, aralkyls, C 1-6 heteroalkyls, substituted C 1-6 heteroalkyls, C 1-6 alkoxy, phenoxy, C 1-6 heteroalkoxy, halo, nitro-, cyano-, hydroxyl-, amino-, carboxy- and trifluoromethyl,

(i) wherein the substituted alkyls, substituted cycloalkyls, substituted aryls and substituted heteroalkyls are independently substituted with the group consisting of halo, alkoxy, nitro, carboxy, amino, alkylamino, hydroxyl and mercapto; and

(ii) wherein a carbon of the heteroalkyls and heteroalkoxy is independently substituted with the group consisting of O, N and S;

comprising:

a) deprotonating a paullone,

b) attaching a blocked bifunctional spacer to form a blocked intermediate, and

c) deblocking said blocked intermediate and reacting the deblocked intermediate with an activated polymer

under conditions sufficient to cause a polymeric conjugate to be formed.
