Abstract

Methods and compositions are provided for detecting biomolecular interactions. The use of labels is not required and the methods can be performed in a high-throughput manner. The invention also provides optical devices useful as narrow band filters.

Claims (12)

1. A method of analyzing or identifying one or more molecules comprising:

(a) contacting a sample comprising the one or more molecules with a colorimetric resonant reflectance optical sensor such that one or more of the one or more molecules become immobilized to the colorimetric resonant reflectance optical sensor;

(b) detecting the one or more molecules immobilized to the colorimetric resonant reflectance optical sensor;

(c) eluting the immobilized one or more molecules from the colorimetric resonant reflectance optical sensor; and

(d) subjecting the one or more molecules to mass spectrometry analysis, wherein one or more molecules are analyzed or identified.

2. The method of claim 1 , wherein the one or more molecules immobilized to the colorimetric resonant reflectance optical sensor are quantified.

The method of claim 1 , wherein the one or more molecules immobilized to the colorimetric resonant reflectance optical sensor are detected by determining a first peak wavelength of the colorimetric resonant reflectance optical sensor prior to contacting the sample with the colorimetric resonant reflectance optical sensor; determining a second peak wavelength after contacting the sample with the colorimetric resonant reflectance optical sensor and comparing the first and second peak wavelengths, wherein a shift in peak wavelength indicates that the one or more molecules of the sample have immobilized to the colorimetric resonant reflectance optical sensor.

4. The method of claim 1 , wherein analyzing or identifying the one or more molecules immobilized to the colorimetric resonant reflectance optical sensor comprises the use of a tag.

5. The method of claim 4 , wherein the tag is biotin, succinimidyl-6-[a-methyl-a-(2-pyridyl-dithio) toluamido] hexanoate (SMPT), dimethylpimelimidate (DMP), or histidine.

6. The method of claim 1 , wherein the one or more molecules are immobilized to the colorimetric resonant reflectance optical sensor by one or more specific binding substances on the surface of the colorimetric resonant reflectance optical sensor.

The method claim 6 , wherein the one or more specific binding substances comprise a nucleic acid, polypeptide, antigen, polyclonal antibody, monoclonal antibody, single chain antibody (scFv), F(ab) fragment, F(ab′) 2 fragment, Fv fragment, small organic molecule, cell, virus, bacteria, biological sample or a combination thereof.

8. The method of claim 1 , wherein the colorimetric resonant reflectance optical sensor is coupled to a microfluidic channel.
