Abstract

The present invention refers to new microspheres including pancreatic enzymes, pharmaceutical compositions containing them, and a process to obtain them. The process here described doesn't involve the use of solvents and proves to be remarkably shorter and efficient than the producing methods of the prior arts. The microspheres obtained, including one or more pancreatic enzymes, one or more hydrophilic low-melting polymers and eventual excipients, have an high enzymatic activity, bio-availability and stability.

Claims (19)

1. A process for the production of microspheres comprising one or more pancreatic enzymes and one or more hydrophilic low-melting polymers, comprising:

a) introducing said one or more pancreatic enzymes and said one or more hydrophilic low-melting polymers or a combination thereof in solid state into a reactor to form a mixture, wherein the temperature of the reactor is below the melting temperature of said one or more hydrophilic low-melting polymers;

b) stirring and heating the mixture to obtain said one or more hydrophilic low-melting polymers in a melted state;

c) cooling the mixture while stirring to obtain solidified microspheres; and

d) recovering said solidified microspheres,

wherein the ratio between said one or more pancreatic enzymes and said one or more hydrophilic low-melting polymers is between 4:1 and 1:1, and said solidified microspheres have a diameter from about 10 pm to about 1500 pm and an enzymatic titer greater than or equal to 90% compared to the enzymatic titer of said one or more pancreatic enzymes before being introduced into said reactor; and the said process is performed in the absence of solvents.

2. The process according to claim 1 , wherein said hydrophilic low-melting polymer has a melting point between 20° C. and 90° C.

3. The process according to claim 1 , wherein said hydrophilic low-melting polymer has a melting point from about 30° C. to about 70° C.

4. The process according to claim 1 , wherein said solid mixture further includes at least one excipient.

5. The process according to claim 1 , wherein said hydrophilic low-melting polymer is chosen from among polyethylene glycol, polyoxyethylene, copolymers of poloxyethylene and polyoxypropylene, and mixtures thereof.

6. The process according to claim 1 , further comprising the step of coating said solidified microspheres with a polymeric film.

7. The process according to claim 1 , wherein the activity of the enzyme in the microsphere has an enzymatic titer greater than or equal to 95% compared to the enzymatic titer of said one or more pancreatic enzymes before being introduced into said reactor.

8. The process according to claim 1 , wherein the mixture is heated at a temperature ranging from about 20° C. to about 90° C. in step b).

9. The process according to claim 1 , wherein the mixture is heated at a temperature ranging from about 30° C. to about 70° C. in step b).

10. The process according to claim 8 , wherein said hydrophilic low-melting polymer has a melting point between 20° C. and 90° C.

11. The process according to claim 9 , wherein said hydrophilic low-melting polymer has a melting point from about 30° C. to about 70° C.

12. A pharmaceutical composition comprising microspheres produced according to claim 1 and one or more excipients.

13. The pharmaceutical composition according to claim 12 , wherein the microspheres comprises one or more pancreatic enzymes.

14. The pharmaceutical composition according to claim 12 , wherein the composition is in form of powder, pellets, capsules, tablets, microtablets, solutions or suspensions.
