Abstract

Method of determining the efficacy of treatment for mycobacterial infection in an individual comprising determining in samples from the individual whether the level of T cells specific for a mycobacterial antigen has decreased after the treatment, thereby determining the efficacy of the treatment.

Claims (13)

1. Method of determining the efficacy of treatment for M. tuberculosis infection in an individual comprising determining in samples from the individual whether the level of T cells specific for an antigen of said M. tuberculosis has decreased after the treatment, wherein said antigen is ESAT-6 or CFP 10, thereby determining the efficacy of the treatment and wherein said determining is by quantification of said T cells as directly ex vivo cells.

2. Method according to claim 1 wherein the T cells are CD4 T cells.

3. Method according to claim 1 comprising detecting the level of the said T cells in a first sample taken before the treatment and in a second sample taken after the treatment.

4. Method according to claim 1 wherein the T cells detected are freshly-isolated T cells from peripheral blood.

5. Method according to claim 1 wherein the level of T cells snecific for said antigen is determined by contacting cells of the sample with said antigen.

6. Method according to claim 5 wherein said determining is performed by detection of a cytokine secreted by the T cells specific for said antigen.

7. Method according to claim 6 in which the cytokine is IFN-γ.

8. Method according to claim 6 in which the cytokine is detected by allowing the cytokine to bind to an immobilized antibody specific to the cytokine and then detecting the presence of the antibody/cytokine complex.

9. Method according to claim 1 comprising determining whether after the treatment a decreased level of T cells specific for said M. tuberculosis antigen continues to be detectable, which decreased level is indicative of latent infection.

10. Method according to claim 1 wherein the level of T cells specific for said antigen is determined by contacting cells of the sample with one or more peptides selected from fragments of said antigen which are capable of recognition by T cells of said individual.

11. Method according to claim 10 wherein said determining is performed by detection of a cytokine secreted by the T cells specific for said antigen.

12. Method according to claim 11 in which the cytokine is IFN-γ.

13. Method according to claim 11 in which the cytokine is detected by allowing the cytokine to bind to an immobilized antibody specific to the cytokine and then detecting the presence of the antibody/cytokine complex.
