Abstract

The present invention relates to compositions and methods for preserving and extracting nucleic acids from saliva. The compositions include a chelating agent, a denaturing agent, buffers to maintain the pH of the composition within ranges desirable for DNA and/or RNA. The compositions may also include a reducing agent and/or antimicrobial agent. The invention extends to methods of using the compositions of the invention to preserve and isolate nucleic acids from saliva as well as to containers for the compositions of the invention.

Claims (54)

1. A method for preserving deoxyribonucleic acid (DNA) contained in sputum comprising the steps of:

a) obtaining sputum from a subject;

b) contacting said sputum with a composition comprising a chelating agent and a denaturing agent, wherein the pH of said composition is greater than 5.0, to form a liquid mixture; and

c) storing said mixture for at least 14 days at room temperature,

wherein said composition stabilizes said DNA for at least 14 days at room temperature.

2. The method of claim 1 , wherein the sputum is saliva.

3. The method of claim 1 , wherein said sputum is from a mammal.

4. The method of claim 1 , wherein said pH is between 7.0 and 10.0, inclusive.

5. The method of claim 1 , with the proviso that said composition does not contain ascorbic acid.

6. The method of claim 1 , wherein said DNA is stable for more than 60 days at room temperature.

7. The method of claim 5 , wherein said DNA is stable for more than 360 days at room temperature.

8. A method for recovering deoxyribonucleic acid (DNA) from sputum comprising the steps of:

a) obtaining sputum from a subject;

b) contacting said sputum with a composition comprising a chelating agent and a denaturing agent, wherein the pH of said composition is greater than 5.0, to form a liquid mixture;

c) storing said mixture for at least 14 days at room temperature, wherein said composition stabilizes said DNA for at least 14 days at room temperature;

d) contacting said mixture with a protease; and

e) recovering said DNA from said mixture.

9. The method of claim 8 , wherein said sputum is saliva.

10. A method for preserving deoxyribonucleic acid (DNA) from a bodily fluid comprising:

i) placing said bodily fluid into a first region of a container;

ii) placing a composition comprising a chelating agent and a denaturing agent, wherein the pH of said composition is greater than 5.0, into a second region of said container, said second region separated from said first region by a barrier;

iii) closing said container;

iv) disestablishing said barrier such that said composition contacts said bodily fluid forming a liquid mixture; and

v) storing said container at room temperature for at least 14 days,

wherein said composition stabilizes said DNA for at least 14 days at room temperature.

11. The method of claim 10 , further comprising recovering said DNA from said mixture.

12. The method of claim 10 , wherein said bodily fluid is sputum.

13. The method of claim 10 , wherein said bodily fluid is saliva.

14. A method for preserving deoxyribonucleic acid (DNA) contained in sputum comprising the steps of:

a) obtaining sputum from a subject;

b) contacting said sputum with a composition comprising a chelating agent, a denaturing agent, and a protease which is proteinase K, wherein the pH of said composition is greater than 5.0, to form a liquid mixture; and

c) storing said mixture for at least 14 days at room temperature,

wherein said composition stabilizes said DNA for at least 14 days at room temperature.

15. The method of claim 14 , wherein said sputum is saliva.

16. The method of claim 14 , wherein said sputum is from a mammal.

17. The method of claim 14 , wherein the pH of said composition is between 7.0 and 10.0, inclusive.

18. The method of claim 14 , with the proviso that said composition does not contain ascorbic acid.

19. The method of claim 18 , wherein said DNA is stable for more than 60 days at room temperature.

20. The method of claim 18 , wherein said DNA is stable for more than 360 days at room temperature.

21. A method for recovering deoxyribonucleic acid (DNA) from sputum comprising the steps of:

a) obtaining sputum from a subject;

b) contacting said sputum with a composition comprising a chelating agent, a denaturing agent, and a protease which is proteinase K, wherein the pH of said composition is greater than 5.0, to form a liquid mixture;

c) storing said mixture for at least 14 days at room temperature, wherein said composition stabilizes said DNA for at least 14 days at room temperature; and

d) recovering said DNA from said mixture.

22. The method of claim 21 , wherein said sputum is saliva.

23. A method for preserving deoxyribonucleic acid (DNA) from a bodily fluid comprising the steps of:

a) placing said bodily fluid into a first region of a container, said first region having a protease which is proteinase K;

b) placing a composition comprising a chelating agent and a denaturing agent, wherein the pH of said composition is greater than 5.0, into a second region of said container, said second region separated from said first region by a barrier;

c) closing said container;

d) disestablishing said barrier such that the composition of step b) contacts said bodily fluid of step a), and

e) storing said container for at least 14 days at room temperature, wherein the resulting mixture stabilizes said DNA for at least 14 days at room temperature.

24. The method of claim 23 , further comprising recovery of said DNA from said mixture.

25. The method of claim 23 , wherein said bodily fluid is sputum.

26. The method of claim 23 , wherein said bodily fluid is saliva.
