Abstract

The present invention relates to a polypeptide having a modified amino acid sequence of 6-phosphogluconate dehydrogenase (hereinafter abbreviated as GND) derived from a microorganism belonging to the genus Corynebacterium , said modification being substitution of the amino acid residue(s) at the position(s) corresponding to the 158th and/or the 361st amino acid(s) of the amino acid sequence shown in SEQ ID NO: 1, and having GND activity; DNA encoding the polypeptide; a recombinant DNA comprising the DNA; a transformant carrying the recombinant DNA; a microorganism carrying the DNA on the chromosome; and a process for producing a useful substance which comprises culturing the transformant or the microorganism in a medium.

Claims (8)

1. An isolated polypeptide having a 95% or more homology to the amino acid sequence of SEQ ID NO: 1 and substitution of the amino acid residue at the position corresponding to the 158th amino acid of the amino acid sequence of SEQ ID NO: 1 by an amino acid residue selected from the group consisting of Ser, Thr, Trp, Cys, Asn, Gln and Tyr, and having GND activity.

An isolated polypeptide having a 95% or more homology to the amino acid sequence of SEQ ID NO: 1, substitution of the amino acid residue at the position corresponding to the 158th amino acid of the amino acid sequence of SEQ ID NO: 1 by an amino acid residue selected from the group consisting of Ser, Thr, Trp, Cys, Asn, Gln and Tyr and substitution of the amino acid residue at the position corresponding to the 361st amino acid of the amino acid sequence of SEQ ID NO: 1 by an amino acid residue selected from the group consisting of Phe, Val, Leu, Ile, Met, Gly, Ala, Pro and Trp, and having GND activity.

3. An isolated polypeptide selected from the group consisting of polypeptides according to the following (a) and (b):

(a) an isolated polypeptide having a modified amino acid sequence of 6-phosphogluconate dehydrogenase (hereinafter abbreviated as GND) isolated from a microorganism belonging to the genus Corynebacterium , said modification consisting of substitution of the amino acid residue at the position corresponding to the 158th amino acid of the amino acid sequence of SEQ ID NO: 1 by an amino acid residue selected from the group consisting of Ser, Thr, Trp, Cys, Asn, Gln and Tyr, and having GND activity; and

(b) an isolated polypeptide having a modified amino acid sequence of GND isolated from a microorganism belonging to the genus Corynebacterium , said modification consisting of substitution of the amino acid residue at the position corresponding to the 158th amino acid of the amino acid sequence of SEQ ID NO: 1 by an amino acid residue selected from the group consisting of Ser, Thr, Trp, Cys, Asn, Gln and Tyr and substitution of the amino acid residue at the position corresponding to the 361st amino acid of the amino acid sequence of SEQ ID NO: 1 by an amino acid residue selected from the group consisting of Phe, Val, Leu, Ile, Met, Gly, Ala, Pro and Trp, and having GND activity,

wherein the GND isolated from the microorganism belonging to the genus Corynebacterium is GND having the amino acid sequence of SEQ ID NO: 1.

4. The isolated polypeptide according to claim 3 , wherein the amino acid residue selected from the group consisting of Ser, Thr, Trp, Cys, Asn, Gln and Tyr is a Ser residue.

The isolated polypeptide according to claim 3 , wherein the amino acid residue selected from the group consisting of Ser, Thr, Trp, Cys, Asn, Gln and Tyr is a Ser residue and the amino acid residue selected from the group consisting of Phe, Val, Leu, Ile, Met, Gly, Ala, Pro and Trp is a Phe residue.
