Abstract

It is intended to provide agents for treating mild cognitive impairment which comprise compounds having an effect of inhibiting the production, secretion, aggregation, and/or accumulation of β-amyloid proteins, prodrugs thereof or salts of the same and inhibit the progress of mild cognitive impairment to Alzheimer's disease using the same.

Claims (14)

1 . A agent for preventing or treating mild cognitive impairment which comprises a compound having an inhibitory effect on production, secretion, aggregation and/or accumulation of β-amyloid protein or a prodrug thereof.

2 . The agent according to claim 1 wherein the compound is a compound having an inhibitory effect on β-secretase.

3 . The agent according to claim 1 wherein the compound is a compound represented by the formula:

wherein, Ar is an optionally substituted aromatic group, x is a divalent C 1-6 aliphatic hydrocarbon group which may contain one or two divalent groups selected from —O—, —S—, —CO—, —SO—, —SO 2 — and —COO—, Y is a divalent C 1-6 aliphatic hydrocarbon group, R 1 and R 2 may be the same or different and each is a hydrogen atom or optionally substituted C 1-6 alkyl, ring a is a benzene ring which may have an additional substituent, and ring B is a 4- to 8-membered ring which may have an additional substituent, or a salt or hydrate thereof.

The agent according to claim 3 wherein the compound is selected from 6-(4-biphenylyl)methoxy-2-[2-(N,N-dimethylamino)ethyl]tetralin, 6-(4-biphenylyl)methoxy-2-(N,N-dimethylamino)methyltetralin, 6-(4-biphenylyl)methoxy-2-(N,N-dipropylamino)methyltetralin, 2-(N,N-dimethylamino)methyl-6-(4′-methoxybiphenyl-4-yl)methoxytetralin, 6-(4-biphenylyl)methoxy-2-[2-(N,N-diethylamino)ethyl]tetralin, 2-[2-(N,N-dimethylamino)ethyl]-6-(4′-methylbiphenyl-4-yl)methoxytetralin, 2-[2-(N,N-dimethylamino)ethyl]-6-(4′-methoxybiphenyl-4-yl)methoxytetralin, 6-(2′,4′-dimethoxybiphenyl-4-yl)methoxy-2-[2-(N,N-dimethylamino)ethyl]tetralin, 6-[4-(1,3-benzodioxol-5-yl)phenyl]methoxy-2-[2-(N,N-dimethylamino)ethyl]tetralin, 6-(3′,4′-dimethoxybiphenyl-4-yl)methoxy-2-[2-(N,N-dimethylamino)ethyl]tetralin, and optical active forms, salts and hydrates thereof.

5 . The agent according to claim 2 wherein the compound having an inhibitory effect on β-secretase is OM99-2.

6 . The agent according to claim 1 wherein the compound is a compound having an inhibitory effect on γ-secretase.

7 . The agent according to claim 1 wherein the compound is a compound having an inhibitory effect on aggregation of β-amyloid protein.

8 . The agent according to claim 7 wherein the compound having an inhibitory effect on aggregation of β-amyloid protein is PTI-00703, ALZHEMED (NC-531), PPI-368, PPI-558 or SKF-74652.

9 . An agent for inhibiting the progression of mild cognitive impairment to Alzheimer's disease, which comprises a compound having an inhibitory effect on production, secretion, aggregation and/or accumulation of β-amyloid protein or a prodrug thereof.

10 . A method for preventing or treating mild cognitive impairment, which comprises administering to a mammal an effective amount of a compound having an inhibitory effect on production, secretion, aggregation and/or accumulation of β-amyloid protein or a prodrug thereof.

11 . A method for inhibiting the progression of mild cognitive impairment to Alzheimer's disease, which comprises administering to a mammal an effective amount of a compound having an inhibitory effect on production, secretion, aggregation and/or accumulation of β-amyloid protein or a prodrug thereof.

12 . Use of a compound having an inhibitory effect on production, secretion, aggregation and/or accumulation of β-amyloid protein or a prodrug thereof, for the manufacture of an agent for preventing or treating mild cognitive impairment.

13 . Use of a compound having an inhibitory effect on production, secretion, aggregation and/or accumulation of β-amyloid protein or a prodrug thereof, for the manufacture of an agent for inhibiting the progression of mild cognitive impairment to Alzheimer's disease.
