Abstract

The present invention relates to an immunoglobulin-binding protein, wherein at least one asparagine residue has been mutated to an amino acid other than glutamine or aspartic acid, which mutation confers an increased chemical stability at pH-values of up to about 13-14 compared to the parental molecule. The protein can for example be derived from a protein capable of binding to other regions of the immunoglobulin molecule than the complementarity determining regions (CDR), such as protein A, and preferably the B-domain of Staphylococcal protein A. The invention also relates to a matrix for affinity separation, which comprises an immunoglobulin-binding protein as ligand coupled to a solid support, in which protein ligand at least one asparagine residue has been mutated to an amino acid other than glutamine.

Claims (17)

1. In an immunoglobulin-binding protein of SEQ ID NO. 1 or SEQ ID NO. 2, capable of binding to regions of an immunoglobulin molecule other than complementarity determining regions (CDR), the improvement consisting of mutating one asparagine residue of the protein having an amino acid sequence of SEQ ID NO 1 or 2 with an amino acid other than glutamine, wherein said mutated asparagine is N23T, which mutation confers an increased chemical stability at alkaline pH-value.

2. The protein of claim 1 , which binds to an Fc fragment and the Fc portion of a full antibody.

3. A multimer comprised of two or more proteins of claim 1 .

4. The multimer of claim 3 , wherein the proteins are linked by elements including up to about 15 amino acids.

5. The multimer of claim 3 , further comprising one or more of the E, D, A, B, and C domains of Staphylococcus protein A.

6. The multimer of claim 3 , comprising three of said proteins.

7. The multimer of claim 3 , further comprising the protein defined by SEQ ID NO. 2.

8. A nucleic acid encoding an immunoglobulin-binding protein of claim 1 .

9. An expression system, which includes the nucleic acid of claim 8 .

10. A matrix for affinity chromatography, comprising a plurality of ligands which include the immunoglobulin-binding protein of claim 1 coupled to a solid support.

11. The matrix of claim 10 , wherein the ligands are coupled to the support by thioether bonding.

12. The matrix of claim 10 , wherein the support is a natural polymer material.

13. The matrix of claim 10 , which selectively binds an immunoglobulin selected from the group consisting of IgG, IgA and IgM.

14. The matrix of claim 10 , wherein the ligands exhibit an increased binding capacity, during two or more separations with intermittent alkaline cleaning, as compared to the ligands which include the protein having an amino acid sequence of SEQ ID NO 1 or 2.

15. The matrix of claim 14 , wherein the cleaning is performed with NaOH and the concentration thereof is up to about 1 M.

16. A nucleic acid encoding a multimer of claim 3 .

17. An expression system, which includes the nucleic acid of claim 16 .
