Abstract

Disclosing a promoter of the protease MDTS9, which is induced by TGF-β, which is involved in the metabolism of extracellular matrix, and in which the expression of the gene thereof is elevated in renal failure model animals, which promoter is useful as a tool for screening an agent for treating and/or preventing chronic renal failure, as well as a method for screening an agent for treating and/or preventing chronic renal failure by selecting an inhibitor of the promoter.

Claims (15)

1 . An isolated polynucleotide molecule having a promoter activity for a polynucleotide encoding the amino acid sequence of SEQ ID NO:2, wherein said polynucleotide molecule comprises a polynucleotide sequence selected from the group) consisting of:

(a) the polynucleotide sequence of SEQ ID NO:17,

(b) a polynucleotide sequence of SEQ ID NO:17 in which 1 to 10 nucleotides are substituted, deleted, and/or inserted,

(c) the polynucleotide sequence consisting of nucleotides 3253 to 5023 of SEQ ID NO:17, and

(d) a polynucleotide sequence consisting of nucleotides 3253 to 5023 of SEQ ID NO:17 in which 1 to 10 nucleotides are substituted, deleted, and/or inserted.

2 . The polynucleotide molecule according to claim 1 , wherein said polynucleotide molecule comprises the polynucleotide sequence of SEQ ID NO:17 or a polynucleotide sequence of SEQ ID NO:17 in which 1 to 10 nucleotides are substituted, deleted, and/or inserted.

3 . The polynucleotide molecule according to claim 1 , wherein said polynucleotide molecule comprises the polynucleotide sequence consisting of nucleotides 3253 to 5023 of SEQ ID NO:17 or the polynucleotide sequence of SEQ ID NO:17.

4 . An expression vector comprising the polynucleotide molecule according to any one of claims 1 to 3 .

5 . A cell transfected with the expression vector according to claim 4 .

A method for analyzing a test compound about whether or not the test compound inhibits the promoter activity of the polynucleotide according to any one of claims 1 to 3 , comprising (1) a step of allowing the test compound to contact with the cell according to claim 5 and (2) a step of detecting the promoter activity.

9 . A method for screening a substance suppressing the expression of a polypeptide of SEQ ID NO:2, comprising the analyzing step by the method according to claim 8 and a step of selecting a substance inhibiting the promoter activity.

11 . A method for producing a pharmaceutical composition for the treatment and/or prevention of chronic renal failure, comprising the analyzing step by the method according to claim 8 and a formulation step.

12 . The polynucleotide molecule according to claim 1 , wherein said polynucleotide molecule consists of the polynucleotide sequence of nucleotides 3253 to 5023 of SEQ ID NO:17 or the polynucleotide sequence of SEQ ID NO:17.
