Abstract

The present invention relates to novel polypeptide variants of factor VII (FVII) or factor VIIa (FVIIa) polypeptides, where said variants comprise an amino acid substitution in position 10 and 32 and where said variants further comprise a sugar moiety covalently attached to an introduced in vivo N-glycosylation site located outside the Gla domain. Such polypeptide variants are useful in therapy, in particular for the treatment of a variety of coagulation-related disorders, such as trauma.

Claims (17)

1. A recombinant Factor VII (FVII) or Factor VIIa (FVIIa) polypeptide variant comprising the amino acid sequence of SEQ ID NO:2, except for substitution with an amino acid selected from the group consisting of P10Q, K32E, T106N and V253N, wherein the polypeptide variant has at least 10% clotting activity compared to the clotting activity of a corresponding wild-type Factor VII or FVIIa polypeptide.

2. The variant of claim 1 , wherein said variant is in its activated form.

3. A method for treating a mammal having a disease or a disorder wherein clot formation is desirable, comprising administering to said mammal an effective amount of the FVII or FVIIa polypeptide variant of claim 1 .

The method of claim 3 , wherein said disease or disorder is selected from the group consisting of a hemorrhage, hemophilia, thrombocytopenia, uncontrolled bleeding, bleeding in a mammal undergoing a transplantation, bleeding in a mammal undergoing resection, gastrointestinal bleeding, variceal bleeding, bleeding due to trauma, and bleeding in surgery.

5. A pharmaceutical composition comprising the variant of claim 1 , and a pharmaceutically acceptable carrier or excipient.

6. A method for treating a mammal having a disease or a disorder wherein clot formation is desirable, comprising administering to said mammal an effective amount of the pharmaceutical composition of claim 5 .

7. The method of claim 6 , wherein said disease or disorder is selected from the group consisting of a hemorrhage, uncontrolled bleeding, bleeding in a mammal undergoing transplantation, bleeding in a mammal undergoing resection, hemophilia, gastrointestinal bleeding, and variceal bleeding.

8. The method of claim 7 , wherein said disease or disorder is hemophilia.

9. The method of claim 7 , wherein said disease or disorder is bleeding in a mammal undergoing a liver transplantation or liver resection.

10. The method of claim 7 , wherein said hemorrhage is a brain hemorrhage.

11. The method of claim 7 , wherein said uncontrolled bleeding is severe uncontrolled bleeding.

12. The method of claim 6 , wherein said disease or disorder is bleeding due to trauma.

13. The method of claim 12 , wherein said trauma is blunt trauma.

14. The method of claim 12 , wherein said trauma is penetrative trauma.

15. The method of claim 6 , wherein said disease or disorder is a bleeding disease or disorder.

16. The method of claim 6 , wherein said disease or disorder is thrombocytopenia.

17. The method of claim 6 , wherein said disease or disorder is bleeding in surgery.
