Abstract

A novel polypeptide which is useful in screening of an agent for improving insulin resistance and an agent for improving glucose metabolism, a polynucleotide encoding the polypeptide, an expression vector comprising the polynucleotide, and a cell transformed with the expression vector are disclosed. The polypeptide is a protein expressed in skeletal muscle. When the protein is overexpressed, incorporation of sugar into a cell is inhibited. A method for screening of an agent for improving insulin resistance and/or an agent for improving glucose metabolism in which the polypeptide is used, and a method for producing a pharmaceutical composition for insulin resistance and/or glucose metabolism improvement comprising a substance obtained according to the method for screening as an active ingredient are also disclosed.

Claims (7)

1. An isolated polypeptide which comprises the amino acid sequence represented by SEQ ID NO:2 or 4, and binds to c-Cbl-associated protein (CAP); or an isolated polypeptide which consists of the amino acid sequence in which 1 to 3 amino acids are deleted, substituted, and/or inserted in the amino acid sequence represented by SEQ ID NO:2 or 4, and binds to CAP.

2. An isolated polypeptide which consists of an amino acid sequence having a sequence identity of 98% or more with the amino acid sequence represented by SEQ ID NO:2 or 4, and is a protein which binds to CAP.

3. An isolated polypeptide consisting of the amino acid sequence represented by SEQ ID NO:2 or 4.

4. A method of screening for an inhibitor of the binding between CAP and the polypeptide according to any one of claims 1 to 3 comprising:

measuring the binding between CAP and said polypeptide in both the presence and absence of a test substance,

wherein a decrease in the binding between CAP and said polypeptide in the presence of the test substance compared to its absence indicates inhibition.

5. The isolated polypeptide according to claim 1 , which comprises the amino acid sequence represented by SEQ ID NO:2 or 4, and binds to c-Cbl-associated protein.
