Mitotic kinesin inhibitors
View Patent ↗The present invention relates to dihydropyrrole compounds that are useful for treating cellular proliferative diseases, for treating disorders associated with KSP kinesin activity, and for inhibiting KSP kinesin. The invention is also related to compositions which comprise these compounds, and methods of using them to treat cancer in mammals.
1. The compound of the formula IV:
or a pharmaceutically acceptable salt or stereoisomer thereof,
wherein:
R 1 and R 2 are independently selected from: H and (C 1 -C 6 )alkyl; and
R 4 is halo.
2. A compound selected from:
(2S)-4-(2,5-Difluorophenyl)-N-[(3R,4R)-4-fluoropyrrolidin-3-yl]-2-(hydroxymethyl)2-phenyl-2,5-dihydro-1H-pyrrole-1-carboxamide,
(2S)-4-(2,5-Difluorophenyl)-N-[(3S,4S)-4-fluoropyrrolidin-3-yl]-2-hydroxymethyl)2-phenyl-2,5-dihydro-1H-pyrrole-1-carboxamide,
(2S)-4-(2,5-Difluorophenyl)-N-[(3R,4R)-4-fluoro-1-methylpyrrolidin-3-yl]-2-(hydroxymethyl)2-phenyl-2,5-dihydro-1H-pyrrole-1-carboxamide,
(2S)4-2,5-Difluorophenyl)-N-[(3S,4S)-4-fluoro-1-methylpyrrolidin-3-yl]-2-(hydroxymethyl)2-phenyl-2,5-dihydro-1H-pyrrole-1-carboxamide,

3. A pharmaceutical composition that is comprised of a compound in accordance with claim 1 and a pharmaceutically acceptable carrier.
