Abstract

A preservation solution for preserving a biological material at a low temperature including one or more polyphenols, and a method for preserving a biological material are provided. The method includes adding the preservation solution to a biological material, cooling the biological material and storing it under appropriate storing conditions. The method can be used for hypothermic preservation or for cryopreservation, including freezing and lyophilization, of any biological material, including cells selected from RBC, WBC, MNC, UCB, HSC and bacteria. A method is also provided for freezing RBCs such that upon thawing, the RBCs include less than 2% free hemoglobin.

Claims (22)

1. A method for preserving a biological material selected from the group consisting of cells, cell aggregates, tissue, organs, natural liposomes and synthetic liposomes, the method comprising:

(a) adding a preservation solution to said biological material, the preservation solution comprising one or more polyphenols and dextran;

(b) freeze drying the biological material; and

(c) storing the biological material under appropriate storing conditions.

2. The method of claim 1 , wherein the one or more polyphenols comprise one or more catechins.

3. The method of claim 2 , wherein the one or more catechins are epigallocatechin gallate (EGCG) catechins.

4. The method of claim 1 , wherein the one or more polyphenols are derived from green tea extract (GTE).

5. The method of claim 1 , wherein the preservation solution does not comprise a significant amount of glycerol.

6. The method of claim 1 , wherein the preservation solution does not comprise a significant amount of DMSO.

7. The method of claim 1 , wherein the preservation solution further comprises trehalose.

8. The method of claim 1 , wherein the preservation is cryopreservation, the preservation solution is a cryopreservation solution, and the freeze drying of step (b) is to a temperature below 0° C.

9. The method of claim 1 , wherein the biological material comprises cells selected from the group consisting of red blood cells (RBC), white blood cells (WBC), mononuclear cells (MNC), umbilical cord blood cells (UCB), hematopoietic stem cells (HSC) and bacteria.

10. A method for preserving a biological material selected from the group consisting of cells, cell aggregates, tissue, organs, natural liposomes and synthetic liposomes, comprising:

(a) adding a preservation solution to the biological material, the preservation solution comprising one or more polyphenols, dextran, and being free from any polyalcohol;

(b) freeze drying the biological material; and

(c) storing the biological material under appropriate storing conditions.

11. The method according to claim 1 , wherein at least 30% of the biological material remains viable.

12. The method according to claim 1 , wherein at least 50% of the biological material remains viable.

13. The method according to claim 1 , wherein at least 75% of the biological material remains viable.

14. The method according to claim 10 , wherein at least 30% of the biological material remains viable.

15. The method according to claim 10 , wherein at least 50% of the biological material remains viable.

16. The method according to claim 10 , wherein at least 75% of the biological material remains viable.
