Abstract

Disclosed is a process for transfecting genetic material into a mammalian cell to alter endogenous properties of the cell. The process comprises designing a polynucleotide for transfection. Then the polynucleotide is inserted into a mammalian vessel such as a tail vein or artery. Prior to insertion, subsequent to insertion, or concurrent with insertion the permeability of the vessel is increased thereby the genetic material is delivered to the parenchymal cell altering endogenous properties of the cell.

Claims (14)

1. A process for delivering a naked non-expressed double strand RNA to an extravascular cell of a mammal, comprising: a) injecting the naked non-expressed double strand RNA into a blood vessel lumen, in vivo; b) increasing the propensity for macromolecules to move through vessel walls and enter the extravascular space; and, c) delivering the naked non-expressed double strand RNA to the extravascular cell outside of the blood vessel.

2. The process of claim 1 wherein the double strand RNA consists of an oligonucleotide of 120 or fewer bases.

3. The process of claim 1 wherein the double strand RNA consists of a polynucleotide of more than 120 bases.

4. The process of claim 1 wherein the non-expressed double strand RNA comprises a sequence that is similar to a gene sequence that is expressed.

5. The process of claim 1 wherein the nucleic acid consists of anti-sense non expressed double strand RNA.

6. The process of claim 1 wherein the propensity for macromolecules to move trough vessel walls and enter the extravascular space is increased by inserting papaverine into the vessel with the naked non-expressed double strand RNA.

7. The process of claim 1 wherein delivery of the naked non-expressed double strand RNA results in decreased expression of a gene.

8. The process of claim 7 wherein the gene is an endogenous gene.

9. The process of claim 7 wherein the gene is a viral gene.

10. The process of claim 1 wherein the vessel consists of a tail vein.

11. The process of claim 1 wherein the parenchymal cell is selected from the group consisting of liver cells, spleen cells, heart cells, kidney cells, prostate cells, skin cells, testis cells, skeletal muscle cells, fat cells, bladder cells, brain cells, pancreas cells, thymus cells, and lung cells.

12. A process for delivering a naked non-expressed double strand RNA to an extravascular cell of a mammal, comprising: injecting the naked nucleic acid into a tail vein, increasing pressure inside the tail vein and delivering the naked nucleic acid to an extravascular cell not within the tail.

The process of claim 12 wherein the parenchymal cell is a cell selected from the group consisting of liver cells, spleen cells, heart cells, kidney cells, prostate cells, skin cells, testis cells, skeletal muscle cells, fat cells, bladder cells, brain cells, pancreas cells, thymus cells, and lung cells.

14. The process of claim 12 wherein the double strand RNA comprises sequence that is similar to an expressed gene sequence.
