Abstract

Modified amino acid compounds useful in the delivery of active agents are provided. The active agents can be peptides, such as rhGH. Methods of administration, such as oral, subcutaneous, sublingual, and intranasal administration, are provided, and methods of preparation of the modified amino acid compound are provided.

Claims (68)

1. The compound

and salts thereof.

2. A composition comprising

(a) an active agent; and

(b) the compound of claim 1 .

3. A composition as defined in claim 2 , wherein said active agent is selected from the group consisting of a pharmacologic agent and a therapeutic active agent.

4. A composition as defined in claim 2 , wherein said active agent is selected from the group consisting of a biologically active agent and a chemically active agent.

5. A composition as defined in claim 4 , wherein said biologically active agent is selected from the group consisting of a peptide, a polysaccharide, a mucopolysaccharide, a carbohydrate, a lipid, a pesticide, and any combination thereof.

6. A composition as defined in claim 5 , wherein said active agent comprises a peptide.

7. A composition as defined in claim 5 , wherein said active agent comprises a polysaccharide.

8. A composition as defined in claim 5 , wherein said active agent comprises a mixture of mucopolysaccharides.

9. A composition as defined in claim 4 , wherein said biologically active agent is a hormone.

10. A composition as defined in claim 8 , wherein said mixture of mucopolysaccharides comprises heparin.

11. A composition as defined in claim 4 , wherein said biologically active agent is selected from the group consisting of human growth hormones; bovine growth hormones; growth releasing hormones; growth hormone-releasing hormones; interferons; interleukin-I; interleukin-II; insulin; heparin, calcitonin; erythropoietin; atrial naturetic factor; antigens; monoclonal antibodies; somatostatin; adrenocorticotropin, gonadotropin releasing hormone; oxytocin; vasopressin; cromolyn sodium; vancomycin; desferrioxamine (DFO); parathyroid hormone, anti-microbials, or any combination thereof.

12. The composition of claim 11 , wherein said biologically active agent comprises insulin.

13. The composition of claim 11 , wherein said biologically active agent comprises heparin.

14. A composition as defined in claim 13 , wherein said heparin comprises low molecular weight heparin.

15. The composition of claim 11 , wherein said biologically active agent comprises calcitonin.

16. A composition as defined in claim 11 , wherein said biologically active agent comprises human growth hormone.

17. The composition of claim 11 , wherein said biologically active agent comprises parathyroid hormone.

18. A dosage unit form comprising a composition as defined in claim 2 ; and

(a) an excipient

(b) a diluent,

(c) a disintegrant,

(d) a lubricant,

(e) a plasticizer,

(f) a colorant,

(g) a dosing vehicle, or

(h) any combination thereof.

19. A dosage unit form according to claim 18 , comprising a tablet, a capsule, or a liquid.

20. A dosage unit form according to claim 19 , consisting of a tablet.

21. A dosage unit form according to claim 19 , consisting of a capsule.

22. The dosage unit form of claim 18 , wherein said active agent is selected from the group consisting of a biologically active agent and a chemically active agent.

23. The dosage unit form of claim 22 , wherein said biologically active agent is selected from the group consisting of a peptide, a polysaccharide, a mucopolysaccharide, a carbohydrate, a lipid, a pesticide, and any combination thereof.

24. The dosage unit form of claim 23 , wherein said biologically active agent comprises a polysaccharide.

25. The dosage unit form of claim 23 , wherein said biologically active agent comprises a peptide.

26. The dosage unit form of claim 23 , wherein said biologically active agent comprises a mixture of mucopolysaccharides.

27. The dosage unit form of claim 26 , wherein said mixture of mucopolysaccharides comprises heparin.

28. The dosage unit form of claim 22 , wherein said biologically active agent is selected from the group consisting of human growth hormones; bovine growth hormones; growth releasing hormones; growth hormone-releasing hormones; interferons; interleukin-I; interleukin-II; insulin; heparin, calcitonin; erythropoietin; atrial naturetic factor; antigens; monoclonal antibodies; somatostatin; adrenocorticotropin, gonadotropin releasing hormone; oxytocin; vasopressin; cromolyn sodium; vancomycin; desferrioxamine (DFO); parathyroid hormone, anti-microbials, or any combination thereof.

29. The dosage unit form of claim 28 , wherein said biologically active agent comprises heparin.

30. The dosage unit form of claim 29 , wherein said heparin comprises low molecular weight heparin.

31. The dosage unit form of claim 28 , wherein said biologically active agent comprises insulin.

32. The dosage unit form of claim 28 , wherein said biologically active agent comprises parathyroid hormones.

33. The dosage unit form of claim 28 , wherein said biologically active agent comprises calcitonin.

34. The dosage unit form of claim 28 , wherein said biologically active agent comprises human growth hormone.

35. A method for preparing a composition, said method comprising mixing:

(a) at least one biologically active agent;

(b) at least one compound as defined in claim 1 ; and

(c) optionally a dosing vehicle.

36. A method as defined in claim 35 , wherein said biologically active agent comprises a peptide.

37. A method as defined in claim 35 , wherein said biologically active agent comprises polysaccharide.

38. A method as defined in claim 35 , wherein said polysaccharide comprises a mixture of muco-polysaccharides.

39. The method of claim 35 , wherein said biologically active agent is selected from the group consisting of human growth hormones; bovine growth hormones; growth releasing hormones; growth hormone-releasing hormones; interferons; interleukin-I; interleukin-II; insulin; heparin, calcitonin; erythropoietin; atrial naturetic factor; antigens; monoclonal antibodies; somatostatin; adrenocorticotropin, gonadotropin releasing hormone; oxytocin; vasopressin; cromolyn sodium; vancomycin; desferrioxamine (DFO); parathyroid hormone, anti-microbials, or any combination thereof.

40. A method of administering a biologically-active agent to an animal in need of said agent, said method comprising administering to said animal a composition as defined in claim 2 .

41. A method for administering a biologically-active agent as defined in claim 40 , wherein said administration is oral.

42. A method for administering a biologically-active agent as defined in claim 40 , wherein said animal is a mammal.

43. A method for administering a biologically-active agent as defined in claim 42 , wherein said mammal is a human.

44. A method for administering a biologically-active agent as defined in claim 40 , wherein said composition is a solid.

45. A method for administering a biologically-active agent as defined in claim 44 , wherein said composition is a tablet or capsule.

46. A method for administering a biologically-active agent to an animal in need of said agent, said method comprising administering orally to said animal a composition as defined in claim 6 .

47. A method for administering a biologically-active agent to an animal in need of said agent, said method comprising administering orally to said animal a composition as defined in claim 8 .

48. A method for administering a biologically-active agent to an animal in need of said agent, said method comprising administering orally to said animal a composition as defined in claim 10 .

49. A method for administering a biologically-active agent to an animal in need of said agent, said method comprising administering orally to said animal a composition as defined in claim 12 .

50. A method for administering a biologically-active agent to an animal in need of said agent, said method comprising administering orally to said animal a composition as defined in claim 13 .

51. A method for administering a biologically-active agent to an animal in need of said agent, said method comprising administering orally to said animal a composition as defined in claim 14 .

52. A method for administering a biologically-active agent to an animal in need of said agent, said method comprising administering orally to said animal a composition as defined in claim 15 .

53. A method for administering a biologically-active agent to an animal in need of said agent, said method comprising administering orally to said animal a composition as defined in claim 16 .

54. A method for administering a biologically-active agent to an animal in need of said agent, said method comprising administering orally to said animal a composition as defined in claim 17 .
